摘要 目的建立高效液相色谱法同时测定柴黄颗粒中5种成分的含量。方法采用Gemini-C18色谱柱(4.6 mm×250 mm,5μm);流动相:乙腈-0.1%磷酸梯度洗脱;流速:1.0 ml/min;柱温:25℃;检测波长:210 nm、280 nm,进样量:10μl。结果柴黄颗粒中黄芩苷在6.096~304.8 ng (r=1.000 0)、汉黄芩苷在3.438~171.9 ng (r=0.999 9)、黄芩素在1.964~98.2 ng (r=0.999 7)、汉黄芩素在0.916~45.8 ng (r=0.999 8)、柴胡皂苷a在21.6~1 080.0 ng (r=0.999 9)范围内线性良好,平均回收率分别为95.16%(RSD=1.65%)、96.59%(RSD=1.30%)、92.33%(RSD=1.92%)、94.87%(RSD=1.42%)、96.77%(RSD=1.73%)。结论所建立的方法具有操作简单、快速、准确度高的优点,可用于柴黄颗粒中5种成分的含量测定。 Objective To develop an HPLC-DAD method for simultaneous determination of 5 components in Chaihuang granules.Methods Gemini-C 18(4.6 mm×250 mm,5μm)column was utilized with acetonitrile-0.1%phosphoric acid as the mobile phase by gradient elution at flow rate of 1.0 ml/min,the column temperature was main-tained at 25℃,the detection wavelength was set at 210 nm and 280 nm,and the injection volume was 10μl.Results Baicalin,wogonoside,baicalein,wogonin and saikosaponin a exhibited a good linear relationship within the ranges of 6.096~304.8 ng(r=1.0000),3.438~171.9 ng(r=0.9999),1.964~98.2 ng(r=0.9997),0.916~45.8 ng(r=0.9998)and 21.6~1080.0 ng(r=0.9999),respectively.The average recoveries of them were 95.16%(RSD=1.65%),96.59%(RSD=1.30%),92.33%(RSD=1.92%),94.87%(RSD=1.42%)and 96.77%(RSD=1.73%),respectively.Conclusion The established method has the advantages of simple operation,rapid anal-ysis and high accuracy,and it is suitable for the determination of 5 components in Chaihuang granules.
