犬细小病毒人工感染模型的建立

摘要 建立犬细小病毒(CPV)人工感染模型,掌握犬细小病毒病的发病规律,为治疗性药品的有效性评价提供对照。选取CPV抗体阴性健康比格犬20只,随机分为A、B、C、D 4个稀释组,每组使用病毒液HA效价分别为2^13,2^10,2^7,2^4,以SD15株CPV作为攻击毒,稀释的病毒液每只犬分3次灌服15 mL。攻毒后,每日观察临床症状并进行评分、采集肛拭子用胶体金抗原检测试纸判断粪便排毒情况;每日采血,全血进行血常规及血液生化检测,血清进行抗体检测,对血象及部分生化指标进行分析。结果显示,以HA价为2^7的病毒液作为发病模型的攻毒液时可使犬在攻毒后的4~5 d发病;发病率100%;临床症状评分逐日增加;在血常规检测中WBC先升高,然后下降;在生化检查中GLOB、tCO2均下降低于正常值;血清检测HI抗体结果由低至高,达到2^10;死亡率高达80%,即C组(2^7)为最适CPV发病模型组。本试验揭示了犬人工感染CPV的发病规律,为CPV治疗性药品有效性评价奠定基础。 In this study,a canine parvovirus artificial infection model was established to master the pathogenesis of canine parvovirus disease,and to provide a control for the evaluation of the effectiveness of therapeutic drugs.Twenty beagles with CPV antibody-negative were randomly divided into four dilution groups,such as A,B,C,and D.The HA values of each group were 2^13,2^10,2^7and 2^4 respectively,the canine parvovirus SD15 strain was used as an attack virus,and the 15 mL diluted virus solution was administered to each dog three times.After the attack,the clinical symptoms were observed and scored daily,and the anal swab was used to determine the detoxification of the feces using the colloidal gold antigen test strip;the blood was collected daily,the blood routine and blood biochemical indexes was detected,and the serum was tested for antibodies.The results showed that when the virus solution with HA price of 2^7 was used as the attacking agent of the disease model,the dog could be infected in 4-5 days after the attack;the incidence rate was 100%;the clinical symptom score increased day by day;in the blood routine test,WBC first elevated,then decreased;GLOB,tCO2 decreased below normal;serum HI antibody results from low to high,reached 2^10;mortality was as high as 80%,the group C(2^7) was the optimal canine parvovirus pathogenesis model group.This article reveals that the pathogenesis of CPV infection in dogs and lays a foundation for the evaluation of the effectiveness of CPV therapeutic drugs.
