摘要
为了研究关岭牛PPAP2B基因mRNA在不同组织中的表达水平及相关功能,试验以关岭牛为实验动物,分别提取心脏、肝脏、小肠、脂肪、大腿肌、背最长肌组织的总RNA,以不同组织的总RNA为模板,逆转录合成cDNA,以GAPDH基因作为内参,应用实时荧光定量PCR技术检测PPAP2B基因mRNA在不同组织中的相对表达量。结果:关岭牛PPAP2B基因mRNA在脂肪中表达量最高,在其他组织中表达量很少甚至几乎不表达。结论:PPAP2B基因可能在牛的脂肪沉积过程中发挥作用。
In order to study the expression level and related functions of PPAP 2 B gene mRNA in different tissues of Guanling cattle,the total RNA of heart,liver,small intestine,fat,thigh muscle and dorsal longissimus muscle was extracted from Guanling cattle.The total RNA of different tissues was used as template,and the reverse transcription cDNA,was synthesized with GAPDH gene as internal reference.The relative expression of PPAP2B gene mRNA in different tissues was detected by real-time fluorescence quantitative PCR.Results:the expression of PPAP 2 B gene mRNA of Guanling cattle was the highest in adipose tissue,but little or no expression in other tissues.Conclusion:PPAP2B gene may play a role in the process of fat deposition in cattle.
