摘要
为了研究促卵泡素β亚基(follicle stimulating hormone,FSHβ)基因在贵州6个地方鸡种中的多态性并进行FSHβ基因生物信息学分析,试验以矮脚鸡、白宜黑鸡、乌蒙凤鸡、高脚鸡、威宁鸡、竹乡鸡为研究对象,分别构建DNA混池,用PCR产物直接测序法检测FSHβ基因多态性,对FSHβ蛋白二级结构、三级结构及基本性质进行生物信息学分析。结果表明:矮脚鸡、白宜黑鸡、乌蒙凤鸡、高脚鸡、威宁鸡和竹乡鸡分别有11,6,7,6,6,3个SNP位点。FSHβ蛋白不稳定且为分泌蛋白,含有信号肽序列,但不是跨膜蛋白。亚细胞定位在分泌通路S上,有一个卷曲螺旋结构(置信值低)和疏水区域,保守结构域在17~123位氨基酸之间。说明贵州6个地方鸡种FSHβ基因具有较高的遗传多样性。
In order to study the polymorphism of Follicle stimulating hormoneβsubunits(FSHβ)gene in 6 local chicken breeds in Guizhou and analyze the biological information of FSHβ gene,Bantam chicken,Baiyi black chicken,Wumeng Crested chicken,Gaojiao chicken,Weining chicken and Zhuxiang chicken were selected as the research subjects in this experiment,respectively constructed DNA mixed pool.FSHβ polymorphism was then sequenced directly with PCR products and the secondary structure,tertiary structure and basic properties of FSHβ protein were analyzed by bioinformatics.The results showed that bantams,Baiyi black chickens,Wumeng Crested chickens,Gaojiao chickens,Weining chickens and Zhuxiang chickens had 11,6,7,6,6,3 SNP respectively.FSHβ protein was unstable and secreted proteins,containing signal peptide sequences,but was not transmembrane proteins.The subcellular localization is in the secretory pathway S,which has a coiled helix structure(low confidence)and a hydrophobic region,and its conserved domains is between 17 to 123 amino acids.The results indicated that the FSHβ genes of the six local chicken species had high genetic diversity.
