摘要 目的:研究连续高压氧(repetitive hyperbaric oxygen,HBO)处理对完全弗氏佐剂(complete Freund′s adjuvant,CFA)诱导的小鼠慢性炎症性疼痛和足底水肿的影响。方法:采用小鼠足底注射CFA诱导小鼠慢性炎症性疼痛。在注射CFA后1 h或3 d每天将小鼠置于高压氧舱中(纯氧,2个大气压,持续5~8 d)1 h,3 h后行为学检测小鼠热痛觉过敏和炎症爪体积;分别在CFA注射后的1、3、7 d取足底皮肤,用Western Blot检测p-c-Jun N末端激酶(c-Jun N-terminal kinase,JNK)、p-细胞外信号调节激酶(extracellular signal regulated kinase,ERK)、瞬时受体电位通道(transient receptor potential channels,TRPV1)的表达。结果:CFA诱导小鼠产生慢性炎症性疼痛和注射爪的水肿;高压氧处理显著缓解热痛觉过敏,并减轻了CFA引起的足部肿胀。而且,CFA诱导足底皮肤中p-ERK、p-JNK和TRPV1在1、3、7 d均显著升高;高压氧处理后的3 d和7 d有效抑制了pERK、pJNK和TRPV1的表达。结论:重复高压氧处理可能通过抑制ERK和JNK的激活以及TRPV1的表达有效缓解慢性炎症性疼痛和局部组织水肿,为临床上采用高压氧镇痛提供了依据。 Objective:To study the effect of repetitive hyperbaric oxygen(HBO)treatment on complete Freund′s adjuvant(CFA)-induced chronic inflammatory pain and paw edema.Methods:Intraplantar injection of CFA was used to induce chronic inflammatory pain.HBO treatment(100%oxygen,2 atmosphere absolute)was given 1 h daily for 5-8 days starting from 1 h or 3 d after CFA injection.Heat hyperalgesia and paw edema were examined 3 h after HBO treatment.The expression of p-extracellular signal regulated kinase(ERK),p-c-Jun N-terminal kinase(JNK),and transient receptor potential channels(TRPV1)in the paw skin was checked in nave mice and HBO-and CFA-treated mice.Results:CFA induced chronic inflammatory pain and paw edema.HBO treatment alleviated heat hyperalgesia and paw edema.CFA also increased the expression of p-ERK,JNK,and TRPV1 in paw skin,which was reduced by HBO treatment.Conclusion:Repetitive HBO treatment may attenuate chronic inflammatory pain and paw edema via inhibition of ERK and JNK activation and TRPV1 expression.The results provide evidence of the antinociceptive effect of HBO on inflammatory pain in clinic.
