酵母β-葡聚糖对脂多糖诱导的绵羊淋巴细胞氧化应激损伤的保护作用 被引量:5

摘要 本试验旨在研究酵母β-葡聚糖对绵羊淋巴细胞抗氧化能力的影响。在建立体外脂多糖(LPS)诱导绵羊淋巴细胞氧化应激模型的基础上,通过在培养液中添加不同浓度[0(对照)、5、10、20、50、100μg/mL]的酵母β-葡聚糖,运用四甲基偶氮唑盐(MTT)比色法测定酵母β-葡聚糖对细胞活性的影响,用生物化学法测定酵母β-葡聚糖对绵羊淋巴细胞抗氧化指标变化的影响。结果表明:0~20μg/mL酵母β-葡聚糖作用于淋巴细胞,细胞存活率无显著变化(P>0.05),当使用50μg/mL酵母β-葡聚糖处理淋巴细胞后,细胞存活率显著降低(P<0.05);在LPS作用于淋巴细胞后,与对照组相比较,丙二醛(MDA)含量极显著增多(P<0.01),超氧化物歧化酶(SOD)和氧化氢酶(CAT)活性显著下降(P<0.05),谷胱甘肽过氧化物酶(GSH-Px)活性极显著下降(P<0.01);添加10μg/mL酵母β-葡聚糖作用6 h后,与LPS组相比较,淋巴细胞内SOD和CAT活性极显著增强(P<0.01),GSH-Px活性显著增强(P<0.05),MDA和ROS含量极显著降低(P<0.01)。综上,LPS可诱导绵羊淋巴细胞氧化应激,酵母β-葡聚糖可抑制LPS诱导的淋巴细胞氧化损伤,对LPS诱导的绵羊淋巴细胞氧化损伤有良好的保护作用,存在剂量依赖效应。 This experiment was conducted to study the effects ofβ⁃glucan from Saccharomyces cerevisiae on the antioxidant capacity of sheep lymphocytes.Based on the establishment of lipopolysaccharide(LPS)⁃in⁃duced oxidative stress in sheep lymphocytes,the effects ofβ⁃glucan on cell viability were determined by MTT assay by adding different concentrations[0(control),5,10,20,50 and 100μg/mL]ofβ⁃glucan to the cul⁃ture medium.The biochemical method was used to determine the effects ofβ⁃glucan on the changes of antioxi⁃dant indexes in sheep lymphocytes.The results showed as follows:β⁃glucan showed no cytotoxicity after being applied to lymphocytes at a concentration of 0 to 20μg/mL,when the concentration ofβ⁃glucan was 50μg/mL,the cell viability decreased significantly(P<0.05);compared with the control group,the malon⁃aldehyde(MDA)content extremely significantly increased(P<0.01),superoxide dismutase(ROS)and cat⁃alase(CAT)activities extremely significantly decreased(P<0.05),and glutathion peroxidase(GSH⁃Px)ac⁃tivity decreased extremely significantly(P<0.01)in lymphocytes treated by LPS.After adding 10μg/mLβ⁃glucan for 6 h,compared with the LPS group,the activity of SOD and CAT in lymphocytes extremely signifi⁃cantly increased(P<0.01),the activity of GSH⁃Px significantly increased(P<0.05),and the contents of MDA and ROS extremely significantly decreased(P<0.01).In conclusions,LPS can induce oxidative stress in sheep lymphocytes.β⁃glucan can inhibit LPS⁃induced oxidative injury of lymphocytes and has a good protec⁃tive effect on LPS⁃induced oxidative injury of sheep lymphocytes,and there is a dose⁃dependent effect.
