摘要
为探讨DHA对胃癌细胞凋亡中MAPKs信号转导通路的诱导作用,运用流式细胞仪技术和MTT法观察和分析细胞凋亡和增殖,并且运用Western blot技术对DHA诱导胃癌细胞的磷酸化水平表达和丝裂原活化蛋白激活ERK1/2水平进行检测。结果发现,DHA可以抑制SGC-7901胃癌细胞增殖,具有浓度和时间依赖性(P<0.05);DHA的作用浓度与SCG-7901细胞凋亡率呈正比关系(P<0.05);同时,随着DHA浓度的升高,caspase-3的表达明显增强,比较有统计意义(P<0.05)。说明,DHA能够对EPK1/2的磷酸化过程进行下调,将凋亡蛋白caspase-3的表达激活,从而对胃癌细胞凋亡发挥诱导作用。
To investigate the effect of DHA on the induction of MAPKs signaling pathway in gastric cancer cell apoptosis,flow cytometry and MTT were used to observe and analyze cell apoptosis and proliferation,and the phosphorylation level expression and mitogen activator protein activation ERK1/2 level of DHA-induced gastric cancer cells were detected by Western blot technique.It was found that DHA could inhibit the proliferation of SCG-7901 gastric cancer cells with concentration and time dependence(P<0.05);the effect concentration of DHA was positively related to the apoptosis rate of SCG-7901 cells(P<0.05);meanwhile,the expression of caspase-3 was significantly enhanced with the increase of DHA concentration,which was statistically significant(P<0.05).It is shown that DHA can down-regulate the phosphorylation process of EPK1/2 and activate the expression of the apoptotic protein caspase-3,thus exerting an induction effect on gastric cancer cell apoptosis.
