摘要
采用超微细胞化学定位技术,在2℃低温处理下,比较研究牛膝菊抗、敏种群叶片间ATP酶的活性定位及低温适应性,结果表明:正常情况下,牛膝菊抗、敏种群叶片的ATP酶均定位于细胞壁,且酶的活性反应无明显差异;2℃低温处理后,抗、敏种群叶片的ATP酶定位位点和活性反应表现出明显差异。其中抗性种群ATP酶起初只定位于细胞壁,在处理4 h后酶活性反应大大增加,细胞膜也出现了致密的线条状分布,在处理8 h时叶绿体边缘也出现酶活性定位点,处理12 h时活性反应下降但结构仍完整;而敏感种群ATP酶仅定位于细胞壁,且定位量没有增加,8 h时出现质壁分离、酶活性完全消失,12 h时细胞超微结构彻底破坏。牛膝菊种群间对低温适应能力不同表现出的抗性差异与ATP酶活性有密切关系。
ATPase activity and low-temperature adaptability of resistant and susceptible populations of Galinsoga parviflora Cav.were studied by ultrastructural cytochemical localization at 2℃low temperature.The results showed that ATPase of resistant and susceptible populations was located at the cell wall,and ATPase activity of them had not significant difference without low temperature treatment.ATPase activity and localization between resistant and susceptible populations at 2℃low temperature had significant difference.After low temperature treatment for 4 h,ATPase of resistant population was localized on cell wall and at plasma membranes.The ATPase activity was intensity.After low temperature treatment for 8 h,ATPase of resistant population was localized on the Chloroplast.After low temperature treatment for 12 h,The ATPase activity of resistant population was still found on cell wall.However,ATPase of susceptible populations was localized only on cell wall and ATPase activity didn’t increase.After low temperature treatment for 8 h,susceptible populations had wall separation.At 2℃,ATPase activity of resistant populations of Galinsoga parviflora Cav.was significantly higher than the that of susceptible species.The low-temperature adaptability of Galinsoga parviflora Cav.was associated with the ATPase activity.
