摘要
扫描电镜观察高脂肪动物样品横切面时,因油脂的覆盖及遮挡,无法观察到肌纤维结构。本文以鸭肉为例,使用不同试剂及不同处理时间对样品进行脱脂处理以去除油脂干扰,比较得到最优方法。实验结果显示,丙酮的脱脂效果优于酒精;不同丙酮时间处理梯度,6~9 h处理为最优。利用该方法,对于高油脂类动物组织,可以在扫描电镜下观察得到清晰的肌纤维超微结构,进而可以快速、准确地对肌肉质构特性进行测定。该方法可以作为评价分析肉品品质的简单方法之一。
When scanning electron microscopy(SEM)was used to observe the cross section of high-fat animal samples,the muscle fibers could not be observed because of the covering and occlusion of oil.In this study,taking duck meat as an example,different reagents and different treatment time were used to degrease the samples in order to remove oil interference,and the best method was obtained.The experimental results showed that the degreasing effect of acetone was better than that of alcohol,and the optimal treatment time was 6-9 hours with different acetone treatment gradients.With this method,clear ultrastructure of muscle fibers of high fat animal tissues can be observed under scanning electron microscope,and then the texture characteristics of muscle can be measured quickly and accurately.This method can be used as one of the simple methods to evaluate and analyze meat quality.
