精胺对雌鹅卵巢生殖激素受体基因表达的影响

摘要 为研究精胺对发育早期雌鹅卵巢生殖激素受体基因表达的影响,本实验分别用禽用生理盐水和5 mg/kg体重的精胺给28日龄雏鹅连续灌胃2周后称重,采集血液和卵巢组织,统计平均日增重、卵巢指数,并利用实时荧光定量PCR检测鹅下丘脑、垂体和卵巢组织中FSHR、LHR、ER1和ER2表达量。结果表明:5 mg/kg体重的精胺灌胃对鹅体重、卵巢重和卵巢/体重比均无显著影响;灌胃精胺组雏鹅卵巢组织LHR和ER2表达量分别是对照组的4.78倍和1.65倍(P<0.05)。上述结果表明,外源性精胺可促进雏鹅卵巢组织ER2和LHR的表达,提示精胺能够通过介导生殖激素受体表达影响鹅的早期卵巢发育。 To study the effect of spermine on reproductive hormone receptor expression in ovary tissues of geese during early development stage, geese aged 28 day-old were fed with spermine(per os) about 2 weeks. The mRNA expression levels of FSHR, LHR, ER1 and ER2 in ovary tissues were examined by real-time PCR. The results showed that the difference in body weight, ovary weight and the ratio of ovary/body weight between 5 mg/kg spermine-treated geese and the control group was not observed(P>0.05). The mRNA expression levels of LHR and ER2 in the ovary of spermine-treated geese was 4.78-and 1.65-fold higher compared with the control group, respectively(P<0.05). In conclusion, spermine upregulates ER2 and LHR transcriptions in ovarian tissues. These results indicate that spermine mediates ovarian early development through mediating hormone receptor gene expressions.
