摘要
为测定AOAC 3MTM Petrifilm^TM菌落总数测试片法、国家标准法(4789.2-2016)与ISO(4833-1:2013)法在食品菌落总数测定方面的相关性及等效性,本文按照ISO 16140-2:2016标准方法测定了香料类辣椒面、坚果类花生和面粉类小麦粉自然样品中的菌落总数对GB法、AOAC法与ISO法分别对实验结果进行标准差分析、线性回归以及Bland-Altman分析法汇总分析。结果表明,与ISO法比较,国标法标准差SD=0.039,相关系数R2=0.95,Bland-Altman分析中测量结果有一个均位95%LoA外。与ISO法比较,AOAC 3MTM PetrifilmTM菌落总数测试片法中,标准差SD=0.037,相关系数R2=0.96,Bland-Altman分析中测量结果中有一个结果位于95%LoA外。结果表明:菌落总数测试片法检测食品中的菌落总数、食品微生物学检验菌落总数测定的国家标准方法(4789.2-2016)与ISO(4833-1:2013)方法具有一致性,故在在实际样品测定时,可以根据环境及耗材需要,选定相应的菌落总数的测定方法。
To determine the correlation and equivalence between the AOAC 3M^TM Petrifilm TM colony total test sheet method,the national standard method(4789.2-2016)and the ISO(4833-1:2013)method in the determination of the total number of food colonies,we determined the total number of colonies in natural samples of spice pepper flour,nut peanut and flour wheat flour according to the ISO 16140-2:2016 standard method.The standard deviation analysis,linear regression and Bland-Altman analysis were carried out by GB method,AOAC method and ISO method.The results show that compared with the ISO method,the standard deviation of GB method SD=0.039,and the correlation coefficient R2=0.95,and the measurement results in Bland-Altman analysis have an average position of 95%.AOAC 3MTM PetrifilmTM standard deviation SD=0.037 and the correlation coefficient R2=0.96 in the total colony count test sheet method compared with the ISO method,one of the measured results in the analysis was located outside 95%LoA.According to the results,the national standard method(4789.2-2016)for the determination of colony count in food and food microbiology was consistent with the ISO(4833-1:2013)method.Therefore,the corresponding method for the determination of colony count can be selected according to the needs of environment and consumables.
