外泌体对老龄小鼠骨髓间充质干细胞衰老影响的初步研究 被引量:1

摘要 目的:初步探讨外泌体(exosome)对老龄小鼠骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)衰老的影响。方法:提取年轻小鼠BMSCs来源的外泌体,作用于老龄小鼠BMSCs,并观察其衰老现象的改变;分别提取年轻小鼠和老龄小鼠BMSCs来源的外泌体,进行基因芯片分析,初步筛选并验证表达差异性较大的微小RNA(microRNAs,miRNAs)在老龄小鼠BMSCs衰老过程中的作用。结果:年轻小鼠BMSCs来源的外泌体作用于老龄小鼠BMSCs后,其衰老情况得到明显改善。芯片分析结果显示,miR-199a-3p和miR-214-3p表达的差异性最为明显,其中老龄小鼠BMSCs转染miR-199a-3p-mimics后,可明显缓解其衰老状态。结论:年轻小鼠BMSCs来源的外泌体可明显减缓老龄小鼠BMSCs的衰老情况,外泌体中所含的miR-199a-3p可能在缓解细胞衰老过程中起到了重要的调节作用,但其具体机制有待进一步研究。 Object:To prelimiarily discuss the effect of exosome on the senescence of bone marrow mesenchymal stem cells(BMSCs)in old mice.Methods:The exosomes derived from young mice BMSCs were extracted to act on the old mice BMSCs,observing the change of aging phenomenon.Furthermore,exosomes from young and old mice BMSCs were extracted and analyzed by microarray.The effect of microRNAs(miRNAs)with different expression on the aging process of old mice BMSCs was preliminarily screened and verified.Results:After the exosomes derived from BMSCs of young mice acting on BMSCs of old mice,the aging situation of the later was obviously improved.The results of microarray analysis showed that the expression differences between miR-199a-3p and miR-214-3p was the most obvious.The BMSCs of old mice transfected with miR-199a-3p-mimics can significantly alleviate their aging state.Conclusion:Exosomes derived from young mice BMSCs can significantly slow down the aging process of old mice BMSCs.MiR-199a-3p in exosomes may play an important regulatory role in the process of cell aging,but its specific mechanism needs further study.
