摘要
铂(IV)具有稳定性高、毒副作用低、易修饰、可口服、克服铂(Ⅱ)药物耐药性等优点,在新型铂类药物开发领域具有广阔的发展前景.炎症是肿瘤的重要特征,对铂(IV)进行功能化修饰,使之在抗肿瘤过程中同时表现出抑制肿瘤相关炎症的特征,对提高药物疗效、降低毒副作用具有重要意义,是铂(IV)药物开发的热点,近年来相关研究工作不断涌现.作者结合自己的工作基础,在总结文献基础上对具有炎症抑制能力的新型铂(IV)药物的研究进展进行了综述.主要包括以下方面:(1)具有抑炎能力的小分子铂(IV)药物;(2)基于药物递送系统的抑炎铂(IV)药物.希望该综述的内容对新型铂类药物研发具有借鉴意义及推动作用.
Platinum(IV)complexes show broad prospects in the development of new platinum drugs for their unique advantages over platinum(Ⅱ)drugs,such as high stability,low toxicity,weak side effects,easy modification,oral administration,overcoming the drug resistance of platinum(Ⅱ)drugs,etc.Inflammation is one of the most important hallmarks of cancers.It is of great significance to prepare platinum(IV)complexes with anti-inflammatory features in curing tumors.They also play important roles in improving drug efficacy and reducing toxic and side effects.A great deal of efforts has been devoted,and many remarkable results have been achieved in recent years.Herein,a review on anti-inflammatory platinum(IV)complexes as antitumor drugs was established based on literatures in and abroad combining with authors′researches.The works were mainly summarized in two aspects:(1)small molecular anti-inflammatory platinum(IV)drugs;(2)anti-inflammatory platinum(IV)drugs based on drug delivery system.It is expected that this review will provide new references for platinum drugs investigations and developments.
