摘要
目的研究异丙酚后处理对大鼠肾缺血再灌注损伤的保护作用,以及对Bcl-2、Bax基因表达的影响。方法 SD大鼠40只,随机分为4组:假手术组(S组)、肾缺血再灌注损伤组(IR组)、异丙酚后处理组(P组)、生理盐水对照组(C组)。IR组大鼠麻醉后,建立肾缺血再灌注损伤模型,缺血时间为45min;P组在IR组基础上于再灌注前5min给予异丙酚20mg/(Kg·h),给药时间为30min;C组仅将P组的异丙酚换为等容量的生理盐水。再灌注24h后,成功麻醉并留取血液标本,处死大鼠留取肾脏组织。生化常规方法检测血清中肌酐(Cr)、尿素氮(UN)的含量;HE染色法观察肾脏的形态学改变;Western Blot检测Bcl-2/Bax的表达水平。结果与S组相比,IR组Cr(315.51±32.66)μmol/L、UN(45.66±11.46)μmol/L含量均显著增高,肾脏组织病变加重,Bcl-2表达减少,Bax增多,差异均具有统计学意义(P<0.01);P组与IR组相比,Cr(283.79±26.26)μmol/L、UN(36.60±6.23)μmol/L含量降低,肾脏组织病变减轻,Bcl-2表达显著增多,而Bax表达明显减少,差异具有统计学意义(P<0.01)。结论异丙酚后处理可以减轻大鼠肾缺血再灌注损伤,其机制可能是通过调节Bcl-2与Bax基因的表达。
Objective To study the protective effect of propofol post-treatment on renal ischemia-reperfusion injury in rats and the effect on Bcl-2 and Bax gene expression.Methods Forty SD rats were randomly divided into 4 groups:sham operation group(S group),renal ischemia-reperfusion injury group(IR group),propofol post-treatment group(P group),and saline control group(C group).After the rats in the IR group were anesthetized,a model of renal ischemia-reperfusion injury was established with an ischemic time for 45 minutes.Group P was given propofol 20 mg/(kg·h)5 minutes before reperfusion on the basis of the IR group for 30 minutes.In group C,propofol was replaced with equal volume of normal saline compared with Group P.After 24 hours of reperfusion,the anesthesia was successfully performed and blood samples were preserved,and the rats were sacrificed and kidney tissues were taken.Routine biochemical methods were used to detect the content of creatinine(Cr)and urea nitrogen(UN)in the serum.HE staining was used to observe the morphological changes of the kidney.Western Blot was used to detect the expression level of Bcl-2/Bax.Results Compared with the S group,the content of Cr(315.51±32.66)μmol/L and UN(45.66±11.46)μmol/L in the IR group were significantly increased.The pathological changes of kidney tissues were aggravated,the expression of Bcl-2 was decreased,and Bax was increased.The differences were statistically significant(P<0.01);Compared with IR group,the content of Cr(283.79±26.26)μmol/L and UN(36.60±6.23)μmol/L in group P were decreased,the renal tissue lesions were alleviated,and the expression of Bcl-2 was increased significantly.The Bax expression was significantly reduced,and the difference was statistically significant(P<0.01).Conclusion Propofol post-treatment can alleviate renal ischemia-reperfusion injury in rats,and its mechanism may be through regulating the expression of Bcl-2 and Bax genes.
