摘要
目的探讨叶酸缺乏对人肺腺癌细胞H1299细胞形态、增殖、凋亡及迁移的影响。方法将人肺癌细胞株H1299分别置于有、无叶酸培养基中(对照组与实验组)连续培养12 d;光学显微镜下观察叶酸对细胞形态变化的影响;采用平板克隆实验、划痕实验评价叶酸对H1299细胞增殖能力及迁移能力的影响;流式细胞术检测2组细胞周期及凋亡变化情况。结果叶酸缺乏可影响H1299细胞形态;与对照组比较,无叶酸培养6 d后H1299细胞增殖能力明显下降,迁移能力减弱,凋亡率显著增加(P<0.05),细胞周期阻滞于S期。结论叶酸缺乏可抑制人肺腺癌细胞株H1299的增殖及迁移能力,造成细胞周期阻滞,诱导细胞凋亡。
Objective To investigate the Effects of folate deficiency on the on the morphology,proliferation and apoptosis of human lung adenocarcinoma cell line H1299 in vitro.Methods The human lung cancer cell line H1299 was cultured in folate and folate-free medium(control group and experimental group)for 12 days.The effects of folate deficiency on cell morphology was observed under light microscope.The effects of folate on proliferation and migration of H1299 cells were evaluated by plate cloning experiment and scratch test.Moreover flow cytometry was used to detect the changes of cell cycle and apoptosis.Results Folate deficiency could affect the morphology of H1299 cells.As compared with those in control group,the abilities of proliferation and migration in H1299 cells were significantly decreased,however the cell apoptosis rate was increased significantly after 6-day culture in experimental group(P<0.05),and the cell cycle was blocked at stage S.Conclusion Folate deficiency can inhibit the the proliferation and migration of human lung adenocarcinoma cell line H1299 to result in the cell cycle arrest and induce cell apoptosis.
