摘要
研究旨在比较分析固醇调节元件结合蛋白-1(Sterol regulatory element-binding protein 1,SREBP-1)基因在湘西黄牛不同组织中的发育性表达规律,以探讨SREBP-1基因与肉牛脂肪代谢的关系。选取湖南德农牧业集团有限公司国家级保种场不同月龄(6月龄、18月龄、30月龄)湘西黄牛各4头,利用实时荧光定量PCR检测肝脏、背最长肌、皮下和腹腔脂肪组织SREBP-1基因的相对表达量。结果表明:①SREBP-1基因在湘西黄牛肝脏、背最长肌、皮下和腹腔脂肪的4个部位中的表达量随月龄增加依次显著增加(P<0.01);②在6月龄时,皮下脂肪SREBP-1基因的表达量显著高于肝脏与腹腔脂肪(P<0.05);③18月龄时,SREBP-1基因在肝脏中的表达量显著高于背最长肌与腹腔脂肪(P<0.05);④30月龄时,SREBP-1基因在肝脏组织中的表达量显著高于其他3个组织(P<0.05),在皮下脂肪中的表达量显著高于背最长肌与腹腔脂肪(P<0.05)。由此可见,SREBP-1基因在湘西黄牛不同部位脂肪组织和肌肉组织中均有表达,且其表达随月龄与部位的不同而不同,存在月龄和部位的差异性。
The objective of study was to observe the SREBP-1 gene expression in different tissues at 6,18,and 30 months age of Xiangxi cattle.For this purpose,4 animals from each age stage were selected to extract the total RNA from liver,longest dorsal muscle,subcutaneous fat and abdominal fat.The relative m RNA expression levels of SREBP-1 gene in four tissues of different ages were detected by real-time quantitative PCR.The results showed as follows: ① The expression level of SREBP-1 gene of liver,longest dorsal muscle,subcutaneous and abdominal fat of Xiangxi cattle increased significantly with the increase of age(P<0.05). ②At the age of 6 months,the relative expression level of SREBP-1 gene in subcutaneous fat was significantly higher than that in liver and abdominal fat(P<0.05).③While at the age of 18 months,the relative expression level of SREBP-1 gene in liver was significantly higher than that in dorsal longest muscle and abdominal fat(P<0.05). ④ At the age of 30 months,the expression level of SREBP-1 gene in liver tissues was significantly higher as compare to other tissues(P<0.05).However,the expression level of SREBP-1 gene in subcutaneous fat was significantly higher than that in dorsal longest muscle and abdominal fat(P<0. 05). These results indicated that SREBP-1 gene expression in adipose tissue and muscle tissue of Xiangxi cattle varies with the age and the tissue type.
