御唐丸对2型糖尿病大鼠空腹血清胰岛素、胰岛素抵抗指数、甘油三酯、总胆固醇水平及胰腺组织胰岛素表达的影响 被引量:2

摘要 目的观察御唐丸对2型糖尿病大鼠空腹血清胰岛素(FINS)、胰岛素抵抗指数(HOMA-IR)、甘油三酯(TG)、总胆固醇(TC)水平及胰腺组织胰岛素表达的影响。方法将150只大鼠随机分为空白对照组20只及造模组130只,造模组以链脲佐菌素(STZ)腹腔注射制作2型糖尿病模型,并选取造模成功的大鼠100只随机分为御唐丸高剂量组、御唐丸低剂量组、糖尿乐胶囊组、盐酸二甲双胍组及模型对照组,每组各20只。御唐丸高、低剂量组分别按2.70、1.35 g/(kg·d)予御唐丸药液灌胃,糖尿乐胶囊组按0.09 g/(kg·d)予糖尿乐胶囊药液灌胃,盐酸二甲双胍组按2.43 g/(kg·d)予盐酸二甲双胍片药液灌胃,空白对照组及模型对照组分别予2 mL/d蒸馏水灌胃,均连续灌胃8周。比较各组大鼠灌胃后血清FINS、HOMA-IR、TG、TC水平及胰腺组织胰岛素表达情况。结果与空白对照组比较,模型对照组灌胃后血清FINS、HOMA-IR、TC及TG水平均明显升高(P<0.05),胰腺组织胰岛素表达明显降低(P<0.05)。与模型对照组比较,糖尿乐胶囊组、盐酸二甲双胍组、御唐丸低剂量组及御唐丸高剂量组灌胃后血清FINS、HOMA-IR、TC及TG水平均明显下降(P<0.05),盐酸二甲双胍组、御唐丸低剂量组及御唐丸高剂量组胰腺组织胰岛素表达明显升高(P<0.05)。与糖尿乐胶囊组比较,御唐丸高剂量组灌胃后血清FINS、HOMA-IR、TC及TG水平均明显降低(P<0.05),胰腺组织胰岛素表达明显升高(P<0.05);御唐丸低剂量组灌胃后仅HOMA-IR及TG水平明显降低(P<0.05)。与盐酸二甲双胍组比较,御唐丸高剂量组灌胃后血清FINS水平明显降低(P<0.05),御唐丸低剂量组TC水平明显升高(P<0.05)。与御唐丸低剂量组比较,御唐丸高剂量组灌胃后血清FINS及TC水平均明显降低(P<0.05),胰腺组织胰岛素表达水平明显升高(P<0.05)。结论御唐丸对糖尿病大鼠有确切疗效,可明显降低大鼠血清FINS及HOMA-IR水平,并提高胰腺组织中的胰岛素的表达,减轻胰岛素抵抗情况,降低TC及TG水平,改善脂质代谢,且有一定的量效关系,高剂量水平可能效果更佳。 Objective To observe the effect of Yutang pill on fasting serum insulin(FINS),insulin resistance index(HOMA-IR),triglyceride(TG),total cholesterol(TC)and insulin expression in pancreas of diabetic rats.Methods 150 rats were randomly divided into blank control group(20 rats)and model group(130 rats).The diabetic model was made by intraperitoneal injection of streptozotocin(STZ)in model group,and the rats were randomly divided into three groups:Yutang pill high dose group,Yutang pill low dose group,Tangniaole capsule group,metformin hydrochloride group and model control group.The high and low dose groups of Yutang pills were given 2.70 and 1.35 g/(kg·d)Yutang pill liquid respectively by gavage.The Tangniaole capsule group were given 0.09 g/(kg·d)of Tangniaole capsule liquid by gavage.The metformin hydrochloride group was given 2.43 g/(kg·d)metformin hydrochloride liquid by gavage.The blank control group and model control group were given equal volume distilled water by gavage.All groups were treated for 8 weeks.The levels of FINS,HOMA-IR,TG,TC and the expression of insulin in pancreas were compared between each group.Results Compared with the blank control group,the levels of FINS,HOMA-IR,TC and TG in the model control group were significantly higher(P<0.05),and the expression of insulin in pancreatic tissue was significantly lower(P<0.05).Compared with the model control group,the levels of FINS,HOMA-IR,TC and TG were decreased significantly in the Tangniaole capsule group,metformin hydrochloride group,Yutang pill low dose group and Yutang pill high dose group(P<0.05).However,the expression of insulin in pancreas tissue of metformin hydrochloride group,Yutang pill low dose group and Yutang pill high dose group increased significantly(P<0.05).Compared with Tangniaole capsule group,the levels of FINS,HOMA-IR,TC and TG in high dose group were significantly lower than those in Tangniaole capsule group(P<0.05),and the expression of insulin in pancreatic tissue was significantly higher than that in Tangniaole capsule group(P<0.05),while the level of HOMA-IR and TG in low dose group was significantly lower than that in Tangniaole capsule group(P<0.05).Compared with metformin hydrochloride group,the FINS level in high dose group was significantly lower than that in metformin hydrochloride group(P<0.05),but the TC level in Yutang pill low dose group was significantly higher than that in metformin hydrochloride group(P<0.05).Compared with Yutang pill low dose group,the levels of FINS and TC in Yutang pill high dose group were significantly lower than those in Yutang pill low dose group(P<0.05),and the expression level of insulin in pancreatic tissue was significantly higher than that in low dose Yutang pill group(P<0.05).Conclusion Yutang pill has definite therapeutic effect on diabetic rats.It can obviously reduce the levels of fins and HOMA-IR,reduce the situation of insulin resistance,reduce the levels of TC and TG,improve lipid metabolism,and improve the expression of insulin in pancreatic tissue.It has a certain dose-response relationship for the treatment of type 2 diabetes,and the effect may be better at high dose level.
