摘要
为贵州地方高粱种质资源在育种中的应用提供科学依据,采用籽粒纵切剖面观察鉴定胚乳性状,改良CTAB法提取DNA等方法,对前期搜集的贵州糯质高粱的GBSSI基因型进行鉴定,并结合分子学方法分析100份糯质材料的wx基因型。结果表明:68份材料为wxa基因型,8份材料为wxc基因型,6份材料为wxd基因型。9份材料含有2种wx基因型,9份材料除已知wx基因型外,还含有野生型等未鉴定基因型。为保持材料wx等位基因多样性,应多株混繁保存。
The GBSSI genotype in glutinous sorghum collected from Guizhou is indentified by observing endosperm traits through kernel longitudinal cutting section and extracting DNA through improved CTAB method,and then wxgenotype of 100 glutinous sorghum accessions is analyzed combining with molecular method to provide the scientific basis for application of local sorghum germplasm resource in breeding in Guizhou.Result:68 glutinous sorghum accessions belong to wxagenotype,8 glutinous sorghum accessions belong to wxcgenotype and 6 glutinous sorghum accessions belong to wxdgenotype.9 glutinous sorghum accessions contain 2 wxgenotypes.In addition to containing known wxgenotype,9 glutinous sorghum accessions also contain wild genotype and unidentified genotype.Multiple plants mixed propagation and preservation pattern should be taken to maintain diversity of wx allele in glutinous sorghum accessions.
