摘要
为白斑狗鱼(Esox lucius)性别发育机制和功能分析研究提供基础资料,通过克隆FOXL2基因cDNA序列,对其进行相关生物信息学分析,并进行荧光定量PCR检测FOXL2基因在白斑狗鱼不同时期各组织的表达情况。结果表明:以白斑狗鱼卵巢cDNA为模板克隆得1 483bp的FOXL2基因序列,其编码292个氨基酸与鲑形目鱼类的FOXL2氨基酸序列同源性较高;FOXL2基因在白斑狗鱼性腺、鳃和脑中均有不同程度的表达,在其他组织中基本无表达,卵巢表达量是精巢表达量的12倍,呈明显的性别二态性。推测FOXL2基因可能与白斑狗鱼卵巢发育和功能维持相关。
The relative bioinformatics of FOXL2 gene are analyzed by cloning cDNA sequence of FOXL2 gene,and then the expression of FOXL2 gene in different tissues at different stage of Esox lucius is detected by fluorescent quantitative PCR to provide the basic data for analyzing and studying of gender development mechanism and function of E.lucius.Result:A 1483 bp sequence of FOXL2 gene is cloned by using cDNA of E.lucius ovary as template,which encoding 292 amino acids,and these amino acid sequences were highly homologous to the FOXL2 amino acid sequence of the salmoniformes.FOXL2 gene is expressed with different degrees in gonad,gill and brain of E.lucius,while is not expressed in other tissues.The expression in ovary is 12 times higher than testicle,which shows an obvious sexual dimorphism.It is speculated that FOXL2 gene may be related to ovarian development and function maintenance in E.lucius.
