摘要
利用体外方法对对羟基苯乙酮的局部安全性进行研究,为化妆品的安全性评估提供依据。所使用的模型或方法包括三维皮肤模型、三维角膜模型、HET-CAM鸡胚绒毛尿囊膜实验、DPRA直接多肽反应实验、h-CLAT人细胞系活化试验等。结果显示:质量分数5%对羟基苯乙酮不会对皮肤模型中的组织活力造成显著影响;三维眼角膜模型实验表明,对羟基苯乙酮的IC50(50%细胞存活时的受试物浓度)为3.1%;当质量分数达到2%时,该物质的鸡胚实验刺激评分为0;DPRA和h-CLAT结果均为阴性。综合上述,对羟基苯乙酮作为化妆品原料在一定使用浓度下,引起皮肤刺激、眼刺激以及皮肤致敏的风险很低。
To provide a basis for safety evaluation of cosmetics,the local toxicological data of p-hydroxyacetophenone was studied by several in vitro methods,including reconstructed skin or cornea-like epithelium models,HET-CAM test method,DPRA and h-CLAT.The results showed that:①even up to mass fraction of 5%p-hydroxyacetophenone did not significantly induce obvious changes of skin tissue viability;②IC50 of p-hydroxyacetophenone in cornea model was 3.1%;③when mass fraction of p-hydroxyacetophenone reached 2%,the vascular hemorrhage score was zero;④the results of DPRA and h-CLAT were all negative.In summary,it can be concluded that p-hydroxyacetophenone can be used in cosmetics with low risk of inducing skin irritation,eye irritation and skin sensitization at a certain use concentration.
