摘要
以钝顶螺旋藻为材料,采用反复冻融法提取藻蛋白粗提物,运用考马斯亮蓝法测定藻蛋白含量。通过钝顶螺旋藻藻蛋白粗提物对DPPH、-OH、·O2-三种自由基的清除能力来评价其体外抗氧化性能,用VC作为对照。试验结果表明:冻融次数6次、冷冻时间6 h、冷冻温度-18℃、融化时间6 h、融化温度37℃时,钝顶螺旋藻藻蛋白的提取率达9.1%;钝顶螺旋藻藻蛋白对DPPH、-OH、·O2-这三种自由基的清除率IC50值分别为1.242、1.196、2.939 g/L,而VC对这三种自由基的清除率的IC50值分别为0.469、0.497、0.474 g/L。虽然钝顶螺旋藻藻蛋白的抗氧化性弱于VC,但其IC50值均小于10 g/L,仍表现出较强的抗氧化性。因此,研究为钝顶螺旋藻藻蛋白作为优质饲料开发提供了参考依据,同时为抗生素饲料的减少使用创造了可能。
Spirulina platensis was used as the material and the crude extracts of algal protein was extracted by repeated freeze-thaw method. The protein content of algal flour was determined by Coomassie Brilliant Blue method. The antioxidant capacity of the crude extracts of algae protein against DPPH,-OH and ·O2-in vitro was evaluated, and VC was used as control. The results showed that the extraction rate of spirulina platensis was 9.1% when the number of freezing and thawing was six times, the freezing time was 6 h, the freezing temperature was -18 ℃, the melting time was 6 h, and the melting temperature was 37 ℃. The three free radicals clearance rates of DPPH,-OH and ·O2- were estimated by the half maximal inhibitory concentration(IC50), and the value were 1.242, 1.196, 2.939 g/L, respectively, while the IC50 value of VC were 0.469, 0.497, and 0.474 g/L, respectively. Although the antioxidant capacity of algal protein from Spirulina platensis was weaker than VC, it’s IC50 value was lower than 10 g/L. It indicated that the algal protein from Spirulina platensis had stronger antioxidant capacity. Therefore, it provides some reference for the developing of Spirulina platensis protein as high quality fodder and it creates possibilities for the reducing use of antibiotic feed.
