摘要
目的探讨血管紧张素转化酶2(ACE2)在结直肠癌中的表达及意义。方法利用Oncomine数据库(http://www.oncomine.org/resource)、人类蛋白质图谱(HPA)数据库(http://www.proteinatlas.org/)、GEPIA数据库(http://gepia.cancer-pku.cn)提取数据,比较结直肠癌组织与正常结直肠组织中ACE2表达,分析ACE2基因表达量与错配修复基因表达量的相关性,同时比较结直肠癌患者ACE2基因高、低表达组生存率。结果结直肠癌组织中ACE2基因表达量较正常结直肠组织明显下调(P<0.05),ACE2蛋白表达较正常结直肠组织明显上调。通过GEPIA数据库分析发现,ACE2基因表达量与错配修复基因MLH1、MSH2、MSH6、PMS1和PMS2表达量均有明显相关性(均P<0.05);结直肠癌患者ACE2基因高、低表达组总生存率及无病生存率比较,差异均无统计学意义(均P>0.05)。结论ACE2蛋白在结直肠癌组织中呈高表达,对结直肠癌患者的生存无明显影响,但与涉及结直肠癌发生的错配修复基因表达关系密切。
Objective To investigate the expression and significance of angiotensin converting enzyme 2(ACE2)in colorectal cancer.Methods Data were extracted from the Oncomine database(http://www.oncomine.org/resource/login.html),human protein atlas(HPA)database(http://www.proteinatlas.org/)and GEPIA database(http://gepia.cancer-pku.cn)to compare the expression of ACE2 in colorectal cancer tissues and normal colorectal cancer tissues.The correlation of the expression of ACE2 gene with the expression of mismatch repair gene,and with the survival of colorectal cancer patients was analyzed.Results ACE2-mRNA expression was down-regulated in colorectal cancer tissues compared with normal colorectal tissues(P<0.05).ACE2 protein expression was up-regulated in colorectal cancer tissues compared with normal colorectal tissues.The expression of ACE2 gene was significantly correlated with the expression of mismatch repair genes MLH1,MSH2,MSH6,PMS1 and PMS2 by GEPIA database analysis(P<0.05).ACE2 gene was not correlated with overall survival rate and disease-free survival rate of colorectal cancer patients(P>0.05).Conclusion ACE2 protein is highly expressed in colorectal cancer tissues and has no significant effect on the survival of colorectal cancer patients,but is closely related to the expression of mismatch repair genes.
