摘要
槲皮素是茶树中一种重要的黄酮醇类化合物,可促进孕烷X受体(Pregnane X receptor,PXR)活性,促进机体排毒。为快速检测分析湖南农业大学长安茶学教学实习基地品种资源库的64个茶树品种绿茶中槲皮素含量,建立了槲皮素含量的高效液相色谱(high performance liquid chromatography,HPLC)分析方法。采用C18色谱柱,流动相组成为0.1%乙酸∶甲醇=50∶50(V∶V),流速1.0 mL/min,柱温30℃,检测波长为360 nm,槲皮素分离度良好,出峰时间较短;在0.76~76.00 mg/L范围内线性关系良好,相关系数大于0.999;检测限(LOQ,S/N=10)为0.152 mg/L;该方法精密度、重复性良好,稳定性相对标准偏差值1.5%;回收率达到97.73%~105.74%。运用该方法对64个不同茶树品种所制绿茶槲皮素含量的差异分析发现,安73、薮北、黄叶水仙、川云809等茶树品种中槲皮素含量较高,分别达3.06 mg/g、3.06 mg/g、2.44 mg/g和2.28 mg/g,是槲皮素等黄酮醇类目标性功能成分的品种选育材料。
Quercetin is an important flavonol compound in tea,which can promote Pregnane X receptor(PXR)activity and accelerate detoxification of the body.In order to quickly detect and analyze the quercetin content in green tea of 64 tea varieties in the variety resource bank of Chang’an tea practice teaching base of Hunan Agricultural University,a high performance liquid chromatography(HPLC)analysis method of quercetin content was established.Using C18 as chromatographic column,0.1%acetic acid and methanol(50:50,V/V)as mobile phase composition,1.0 ml/min flow rate,30℃column temperature,detection wavelength at 360nm,quercetin could be effectively separated and the retention time was short;in the concentration from 0.76 to 76 mg/L was a good linear relation,the correlation coefficient was more than 0.999;the quantitative limit(LOQ,S/N=10)was 0.152 mg/L,the method had good precision and repeatability with a relative standard deviation of 1.5%;the recovery rate for quercetin was 97.73%~105.74%.Using this method for detection the difference quercetin contents of green tea processed from 64 varieties,the results showed that An73,Soubei,Huangyeshuixian,and Chuanyun 809 contained higher quercetin contents,reached 3.06 mg/g,3.06 mg/g,2.44 mg/g,and 2.28 mg/g,respectively,they could be used as breeding materials of tea varieties for target functional components of quercetin and other flavonols.
