摘要
目的观察平喘颗粒对哮喘大鼠肺组织树突细胞(DCs)CD80、CD86、MHCⅡ表达的影响。方法将40只Wistar大鼠随机分为空白组、哮喘组、自噬抑制剂组、平喘颗粒组,每组10只。除空白组外,其余各组均注射卵清蛋白(OVA)致敏液造成慢性哮喘动物模型,空白组则以生理盐水代替OVA激发。空白组、哮喘组给予生理盐水灌胃;自噬抑制剂组在第2次致敏后第4天开始给予60μg/g氯喹腹腔注射,激发阶段则每次激发前1 h给予氯喹腹腔注射;平喘颗粒组给予中药药液灌胃,时间同自噬抑制剂组。取各组大鼠肺组织切片,光镜下检测肺组织炎性浸润情况、DCs浸润情况,采用流式细胞仪检测DCs表面CD80、CD86、MHCⅡ的表达水平。结果与空白组比较,哮喘组大鼠肺组织中可见明显的炎性细胞浸润灶,DCs浸润数量增多(P<0.05);与哮喘组比较,自噬抑制剂组和平喘颗粒组肺组织炎性浸润情况及DCs浸润数量均有不同程度的减轻(P<0.05)。与空白组比较,哮喘组DCs表面CD80、CD86、MHCⅡ的表达水平升高(P<0.05);与哮喘组比较,自噬抑制剂组和平喘颗粒组DCs表面CD80、CD86、MHCⅡ的表达水平降低(P<0.05);与自噬抑制剂组比较,平喘颗粒组DCs表面CD80、CD86、MHCⅡ的表达水平相当(P>0.05)。结论平喘颗粒可以通过降低哮喘大鼠肺组织DCs表面CD80、CD86、MHCⅡ的表达水平,改善哮喘大鼠肺组织中的炎症反应及DCs浸润情况,抑制哮喘的发生。
Objective:To observe the effect of Pingchuan Granule on the expression of CD80,CD86 and MHCⅡin dendritic cells(DCs)of lung tissue of asthmatic mice.Methods:40 Wistar mice were randomly divided into 4 groups:the blank group(group A),asthma group(group B),autophagy inhibitor group(group C)and Pingchuan Granule group(group D),10 mice in each.Groups B,C and D were injected with ovalbumin(OVA)sensitizing solution to cause chronic asthma animal models,while group A was stimulated with physiological saline instead of OVA.The blank group and asthma group were administrated with saline.In autophagy inhibitor group,60μg/g chloroquine(CQ)was injected intraperitoneally on the 4 th day after the second sensitization and CQ was intraperitoneally injected 1 h before each challenge in the challenge phase.Pingchuan Granule group was given intragastric administration of Chinese medicine liquid for the same time as autophagy inhibitor group.The inflammatory infiltration and DCs infiltration of lung tissue sections were detected under light microscope.The expression levels of CD80,CD86 and MHCⅡon the surface of DCs were detected by flow cytometry.Results:Compared with the blank group,the lung tissue of asthma group mice showed obvious inflammatory cell infiltration and the number of DCs infiltration increased(P<0.05).Compared with asthma group,the inflammatory infiltration and the number of DCs infiltration in the lung tissue of autophagy inhibitor group and Pingchuan Granule group were all reduced in varying degrees(P<0.05).Compared with blank group,the expression levels of CD80,CD86 and MHCⅡon the surface of DCs in asthma group were increased(P<0.05).Compared with asthma group,those in autophagy inhibitor group and Pingchuan Granule group were all reduced(P<0.05).Compared with autophagy inhibitor group,those in the Pingchuan Granule group were equivalent(P>0.05).Conclusion:Pingchuan Granule can inhibit the occurrence of asthma by reducing the expression levels of CD80,CD86 and MHCⅡon the surface of DCs,and improve the inflammation and DCs infiltration in lung tissues of asthmatic mice.
