白藜芦醇抗紫外损伤的初步研究 被引量:1

摘要 目的:探索白藜芦醇(RES)干预紫外损伤的有效途径,为研究其药理活性提供新思路。方法:以400 J·m-2的剂量建立紫外(主要是UVC)损伤HeLa细胞的模型;在UV损伤细胞的前、中、后期加入RES进行干预,筛选干预效果最佳的处理方式;对筛选的最佳结果进行细胞水平和分子水平的初步评估。结果:RES干预紫外损伤的最佳方式为与紫外同时处理30 min,最佳处理浓度为50μM,存活率提高了59.5%。RES能通过上调OGG1的表达和减少脂质过氧化产物(MDA)的堆积来抵抗紫外造成的氧化损伤。结论:与紫外同时处理细胞时,RES能显著减轻紫外对细胞的损伤,提示白藜芦醇具有出色的抗紫外损伤能力,推测可将其应用于防晒和护肤产品的研发。 Objective:To explore an effective way for resveratrol(RES)to resist UV damage,and to provide a new idea for studying its pharmacological activity.Methods:The HeLa cells were damaged by UV(mainly UVC)at the dose of 400J·m-2.RES was added before,during and after UV-damaged cells for intervention,and the best treatment was selected.The optimal screening result was initially evaluated at the cellular and molecular levels.Results:The best way to resist UV damage was to treat with RES at the same time for 30min.The best treatment concentration of RES was 50μM and the survival rate was increased by 59.5%.RES resisted the oxidative damage caused by UV by up-regulating the expression of OGG1 and reducing the accumulation of lipid peroxidation product(MDA).Conclusion:When the cells were treated with UV at the same time,RES could significantly reduce the damage caused by UV.That suggests RES has excellent to resisit UV damage and presumably can be applied to the research and development of sunscreen and skin care products.
