基于高通量芯片分析早发性精神分裂症差异表达基因 被引量:2

Analysis on differential expression genes of early-onset schizophrenia based on high-throughput microarray

摘要 目的比较早发性精神分裂症与健康人群之间基因表达差异,探究精神分裂症发病机制。方法 GEO数据库获取基因芯片数据,通过R工具进行差异表达基因分析,使用DAVID工具进行GO富集和通路分析,利用STRING数据库构建蛋白相互作用网络。结果鉴定到314个显著差异表达基因,包括221个显著上调控基因和93个显著下调控基因,基因本体GO显著富集到20个条目,主要涉及g蛋白偶联受体信号通路,嗅觉感知,磷脂酰丝氨酸酰链重构,磷脂代谢过程,钠离子通道调控激活,神经肽绑定等。KEGG通路显著富集结果主要涉及嗅觉信号传导,醚脂类代谢,长期抑制(LTD),甲状腺激素合成等6个通路。蛋白互作网络分析获得5个核心基因,模块分析结果显示得分最高的模块主要与神经肽绑定和刺激神经组织配体受体交互信号通路相关。结论通过生物信息学方法获得了早发性精神分裂症与健康人的差异表达基因,阐释了早发性精神分裂症发病机制的相关基因及其表达特征,为早发性精神分裂症的诊断和治疗提供理论基础。 Objective To identify the differentially expressed genes(DEGs)between early-onset schizophrenia patients and the healthy controls,and to elucidate the potential pathogenesis of schizophrenia.Methods Gene expression chip was downloaded from GEO database.The R software limma package was used to identify the significant differentially expressed genes.Then,DAVID online was used to perform the gene ontology and pathway enrichment analysis.STRING Online was used to integrate the protein-protein interaction network.Results A total of 314 genes with significantly differential expressions were identified,of which 221 genes were significantly up-regulated and 93 genes were significantly down-regulated.The GO enrichment was 20 categories,mainly involved G-protein coupled receptor signaling pathway,sensory perception of smell,phosphatidylserine acyl-chain remodeling,phospholipid metabolic process,sodium channel regulator activity and neuropeptide binding etc.The KEGG enrichment showed that the DEGs mainly involved 6 pathways,like Olfactory transduction,Ether lipid metabolism,Long-term depression,Thyroid hormone synthesis.Five genes were the hub DEGs with the highest degree Module analysis showed that the most significant DEGs were mainly related to Neuroactive ligand-receptor interaction and neuropeptide binding.Conclusion The DEGs between early-onset schizophrenia patients and the healthy controls have been screened by bioinformatics methods,and gene and expression features of early-onset schizophrenia pathogenesis have been explained,which may provide the theoretical basis for the diagnosis and treatment of early-onset schizophrenia.
