摘要
为了研究SVP(SHORT VEGETATIVE PHASE)基因在洋葱花芽分化和发育中的作用,本研究以洋葱“70-1”为试材,克隆得到了1个SVP同源基因,命名为AcSVP,并采用荧光定量PCR对该基因的表达模式进行了分析,结果表明:AcSVP包含1个741 bp的开放阅读框,编码246个氨基酸。氨基酸同源比对结果显示,AcSVP的氨基酸序列与水仙的相似性较高。系统进化结果也显示洋葱的AcSVP与水仙的亲缘关系较近。实时荧光定量分析表明,AcSVP在洋葱未开放的花蕾中表达量最高,在花薹抽出前的叶中次之,在花中的表达量最低。根据以上研究结果推测,AcSVP可能参与调控洋葱的花芽分化。
In order to explore the role of SVP(SHORT VEGETATIVE PHASE),which is a key factor for plant flowering process and plays an important role in flower bud differentiation and plant development of onions,we cloned a homologous gene,named AcSVP,from the Allium cepa L.“70-1”,and used real-time quantitative PCR to analyze the gene expression pattern.The results showed that AcSVP contained a 741 bp open reading frame encoding 246 amino acids.Amino acid homology alignment showed that the amino acid sequence of AcSVP was similar to that of narcissus.The phylogenetic tree clearly showed that the gene shared a closer relationship with narcissus.Real-time quantification expression analysis demonstrated AcSVP had the highest expression in unopened flower buds,followed by leaves before flowering,and the lowest in flowers.These results suggested that AcSVP may be involved in the regulation of flower bud differentiation of onions.
