Development of LAMP and Its Application in Diagnosis of Foot-and-mouth Disease

环介导等温扩增技术(Loop-mediated Isothermal Amplification Method,LAMP)是一门新兴的基因扩增技术,具有操作简单、快速、高特异性、高敏感性、不需要特殊检测设备等特点。随着分子生物学技术的发展,LAMP也得到了长足的发展:多重引物的设计和与ELISA的联合使用使得能够同时检测多种病原以及与实时荧光技术的配合可以实现实时监测反应的过程。LAMP检测技术也已广泛用于临床疾病的诊断、病原微生物,包括流行性细菌或病毒等的定性检测及动物物种和胚胎的性别鉴定,此外它也应用于口蹄疫这种人畜共患的急性传染病的检测中。从简单的LAMP到多重LAMP,再到多重荧光检测以及与其他技术的连用,使得口蹄疫的检测方法不断进步。简述了LAMP的相关原理、发展和应用以及在口蹄疫病毒检测中的应用等研究进展。

The Loop-mediated isothermal amplification method(LAMP)is an emerging gene amplification technology.It has the characteristics of simple operation,fast operation,high specificity,high sensitivity and no need for special detection equipment.With the development of technology,LAMP has not only been fully developed.The design of multiple primers and the combination with ELISA enable the simultaneous detection of multiple pathogens and the cooperation with real-time fluorescence technology to real-time monitoring of the reaction process.LAMP detection technology has also been widely used for the diagnosis of clinical diseases,qualitative detection of pathogenic microorganisms including epidemic bacteria or viruses,and gender identification of animal species and embryos.In addition,it is also used in diagnosis of foot-and-mouth disease,a zoonotic acute infectious disease.From simple LAMP to multiple LAMP to multiple fluorescence detection and in conjunction with other technologies,the detection of foot-and-mouth disease is constantly evolving.We describe the related principles,development and application of LAMP and its application in the detection of foot-and-mouth disease virus in this article.
