摘要
目的探讨姜黄素诱导活化的大鼠肝星状细胞凋亡作用方法培养大鼠肝星状细胞HSC-T6细胞,经TGF-P活化后加入不同浓度姜黄素处理24 h后收集细胞。分别检测细胞增殖和细胞凋亡情况;测定JAK2和STAT3的mRNA以及蛋白表达情况t结果CCK-8法和流式细胞术结果显示,姜黄素降低活化HSC-T6细胞的存活率,促进其细胞凋亡,并呈浓度依赖效应;荧光定量聚合酶链式反应和蛋白印迹实验结果显示,与对照组比较,姜黄素组JAK2和STAT3的mRNA以及蛋白均有下降,且与加入的姜黄素浓度成反比结论姜黄素可以通过下调JAK/STAT信号转导通路的表达来促进活化的大鼠肝星状细胞凋亡,对大鼠肝纤维化起到明显的保护作用。
Objective The apoptosis of rat hepatic stellate cells induced by curcumin and its related mechanisms.Methods HSC-T6 cells were cultured and activated by TGF-P,then treated with curcumin for 24 hours.Cell proliferation and apoptosis were measured separately;m RNA and protein expression of JAK2 and STAT3 were measured.Results The results of CCK-8 assay and flow cytometry showed that curcumin reduced the survival rate of activated HSC-T6 cells and promoted its apoptosis,and showed a concentration-dependent effect.The results of fluorescence quantitative polymerase chain reaction and Western blot showed that compared with the control group,the mRNA and protein of JAK2 and STAT3 in the curcumin group decreased,and it was inversely proportional to the concentration of curcumin added.Conclusions Curcumin can promote the apoptosis of activated rat hepatic stellate cells by dow n-regulating the expression ofJA K/ST AT signal transduction pathway,which has obvious protective effect on rat liver fibrosis.
