10种中药单体改善脂毒性研究 被引量:1

摘要 目的通过评价10种中药单体的抗脂毒性活性,筛选获得可以改善脂肪肝的中药。方法体外培养HepG2细胞,用油酸(OA)和棕榈酸(PA)构建脂毒性模型,选取10种中药活性单体对细胞进行药物干预:酶法检测细胞内甘油三酯含量,Bodipy染色法观察胞内的脂滴;LDH方法检测乳酸脱氢酶释放率,MTT法测定细胞存活率,利用Hochest染色检测细胞死亡情况,综合分析药物干预前后肝细胞脂质蓄积及死亡的情况变化,通过模型筛选出对OA诱导的脂质沉积及对PA诱导的肝细胞损伤具有保护作用的活性单体。结果栀子苷(40μM),升麻素(40μM),黄芪甲苷(100μM)可以降低由OA诱导的脂质沉积,对PA诱导的细胞凋亡具有保护作用,数据均具有统计学意义(P<0.05)。结论为改善脂毒性功能传统中药的开发提供理论基础。 Objective By evaluating the antilipotoxicity of 10 traditional Chinese medicine monomers,the Chinese medicines that can improve fatty liver disease were screened.Methods HepG2 cells were cultured in vitro,and a lipotoxicity model was constructed with oleic acid(OA)and palmitic acid(PA).10 kinds of traditional Chinese medicine active monomers were used for drug intervention:enzymatic detection of intracellular triglyceride content,Bodipy Staining method was used to observe intracellular lipid droplets.LDH method was used to detect the release rate of lactate dehydrogenase.MTT method was used to determine the cell survival rate.Hochest staining was used to detect cell death comprehensive analysis of changes in liver cell lipid accumulation and death before and after drug intervention,Active monomers with protective effect on OA-induced lipid deposition and PA-induced liver cell damage were screened out by the model.Results Gardenoside(40μM),Cimifugin(40μM),Astragaloside(100μM)reduced the TG content induced by OA and protected against PA-induced apoptosis.The data were statistically significant(P<0.05).Conclusion This paper provides a theoretical basis for the development of traditional Chinese medicine to improve fatty liver function.
