H9N2禽流感病毒对鸡鸭肥大细胞及组胺含量的影响 被引量:5

Effect of H9N2 avian influenza virus on mast cells and histamine in chickens and ducks

摘要 通过观察鸡鸭肥大细胞(MC)分布、数量、脱颗粒以及其释放组胺的情况,以了解低致病性禽流感(LPAI)对不同禽类的炎性损伤影响,本实验选用52 d黑羽乌骨鸡24只和52 d樱桃谷鸭24只,按种类随机分成4组,每组6只,以0 d、3 d、6 d、9 d作分组标记。0 d作为阴性组,静脉注射0.2 m L PBS,3 d、6 d、9 d作为阳性组,静脉注射107.0EID50/0.2 m L H9N2 AIV。按分组标记取样后,通过甲苯胺蓝染色观察MC情况,并用组胺ELISA试剂盒检测外周血中组胺的含量。结果显示,感染AIV后,鸡出现精神沉郁,食欲减退,胸腺和肺肿大,胸腔有积液,肺出血及大量中性粒细胞等情况;鸭出现胸腔积液,肺肿大,肺组织出血和中性粒细胞等情况。感染AIV后,鸡、鸭在肺、气管、肠、法氏囊和胸腺组织中的MC数量极显著升高(P<0.01),脾组织中的MC数量显著升高(P<0.05)。鸡、鸭外周血中的组胺在感染病毒后均显著升高(P<0.05)。表明,低致病性H9N2 AIV可引起不同禽类的炎症损伤,并可导致鸡、鸭的MC增多以及组胺含量升高。 Order tolearn the effect of low pathogenic avian influenza on inflammatory injury of different birds by observing the distribution,number,degranulation and histaminerelease of mast cells(MC)in chickens and ducks.2452-day-aged black silky fowl chicken and 2452-day-aged cherry valley duck were divided into four groups.Six animals in each group were randomly selected and classified according to 0 day,3 days,6 days and 9 days.These animals were injected with 0.2 m L PBS on 0 day,then animals were injected with 107.0 EID50/0.2 m L H9 N2 subtype avian influenza virus.The MC were observed by toluidine staining and histamine in peripheral blood was detected by ELISA.The results showed that:(1)After infected with the virus,the chickens were showed de-pressed spirit,anorexia,swelling of thymus and lungs,effusion in the chest,obvious red cells in the alveolus cavity,a large number of neutrophils in the lung tissue,red cellsand exudates in the tracheal cavity,pleural effusion in the ducks,pulmonary enlargement,hem-orrhage and a large number of neutrophils in the lung tissue;(2)After infected with the virus,the number of MC in lung,trachea,intestine,bursa of fabricius and thymus increased significantly(P<0.01),and the number of MC in spleen increased significantly(P<0.05);(3)After infected with the virus,histamine in peripheral blood of chickens and ducks was increased significantly(P<0.05).In Conclusion,low pathogenic avian influenza H9 N2 virus can cause inflammatory damage in different birds,and increase MC and histamine content in chickens and ducks.
