摘要
目的对2014-2018年云南省文山州从手足口病病例中分离的柯萨奇病毒A10型(coxsackievirus A10,CV-A10)完整VP1区基因(894bp)进行基因特征分析。方法用486/488和487/489引物扩增CV-A10VP1区5′端和3′端基因,用Sequencher 4.0软件对序列进行拼接和编辑,得到CV-A10VP1区全序,并将测得的序列与NCBI数据库中的序列进行比对,构建系统进化树,进行基因特征及分子流行病学分析。结果2014-2018年云南省文山州手足口病实验室监测系统从2774份标本中分离到197株肠道病毒阳性标本,病毒分离率为7.10%(197/2774),其中检出8株CV-A10。基于VP1分型,8株毒株序列均为基因型C,其中4株为分支1;2株为分支2;2株为分支3。8株毒株VP1序列的核苷酸一致性为93.07%~94.10%,氨基酸一致性为97.74%~98.41%,与原型株Kowalik(基因库编号:AF081300)的核苷酸一致性为76.01%~76.65%,氨基酸一致性为91.78%~92.26%。结论2014-2018年云南省文山州分离的CV-A10毒株均为基因型C,并可分为3个进化分支,应加强对CV-A10的持续监测和基因特征分析。
Objective To study the entire VP1gene sequences(894bp)of coxsackievirus A10(CV-A10)isolated from hand,foot and mouth disease(HFMD)cases from 2014to 2018in Wenshan prefecture of Yunnan province of China.Methods The 5′and 3′fragments of the CV-A10VP1gene was amplified by 486/488 and 487/489primer pairs and was spliced and edited by using Sequencher 4.0software to get the entire VP1 sequence.The sequences were compared with those downloaded from the NCBI nucleotide sequence database,the phylogenetic tree was constructed and the genetic characteristics were analyzed.Results During the 5 years study period,a total of 197strains of enteroviruses were isolated from 2774samples collected from the Wenshan prefecture HFMD surveillance system,with an isolation rate of 7.10%(197/2774),of which,eight strains were CV-A10.All the eight strains were grouped as genotype C based on the VP1sequencing.Of which,four strains were grouped in cluster 1,two in cluster 2,two in cluster 3.The nucleotide identity among eight strains was 93.07%-94.01%,the amino acid homology was 97.74%-98.41%,and the nucleic acid and amino acid homology between the Wenshan prefecture CV-A10and the prototype strain Kowalik(AF081300)were 76.01%-76.65%and 91.78%-92.26%.Conclusions All CV-A10virus strains isolated in Wenshan prefecture of Yunnan province of China from 2014to 2018were genotype C and could be divided into three clusters.
