miR-141和lncRNA-H19在宫颈癌中的表达及临床意义 被引量:7

摘要 目的:检测宫颈癌组织中微小RNA-141(miR-141)与长链非编码RNA-H19(lncRNA-H19)表达情况,并探究其临床意义。方法:选取2013年9月至2014年12月本院确诊收治的宫颈癌患者62例,取宫颈癌组织及癌旁组织;取血清检测肿瘤指标鳞状细胞癌抗原(SCCA)。利用实时定量PCR(qRT-PCR)法检测宫颈癌及癌旁组织中miR-141、lncRNA-H19表达情况,采用全自动化学发光免疫分析仪检测患者血清SCCA水平。结果:与癌旁组织相比,宫颈癌组织中miR-141和lncRNA-H19表达水平显著升高;宫颈癌组织中miR-141表达与患者临床分期、血清SCCA水平显著相关;宫颈癌组织中lncRNA-H19表达与患者有无淋巴结转移、血清SCCA水平显著相关;Pearson分析发现,宫颈癌组织中miR-141和lncRNA-H19表达水平显著正相关;miR-141低表达患者3年总生存率为37.50%,显著高于miR-141高表达者的8.00%;lncRNA-H19低表达患者3年总生存率为41.67%,显著高于lncRNA-H19高表达者的13.16%。结论:miR-141和lncRNA-H19在宫颈癌组织中的表达显著升高,与宫颈癌疾病严重程度有关,可能作为判断宫颈癌患者预后的生物指标。 Objective:To detect the expression of microRNA-141(miR-141)and long non-coding RNA-H19(lncRNA-H19)in cervical cancer and explore their clinical significance.Methods:62 patients with cervical cancer admitted to our hospital from September 2013 to December 2014 were selected.The tissues of cervical cancer and adjacent tissues were collected.Serum samples were taken to detect the tumor marker,squamous cell carcinoma antigen(SCCA).Quantitative Real-time PCR(qRT-PCR)was used to detect the expression level of miR-141 and lncRNA-H19 in ovarian cancer.Automatic chemiluminescence analyzer was used to detect the level of serum SCCA.Results:Compared with adjacent tissues,the expression levels of miR-141 and lncRNA-H19 in cervical cancer tissues increased significantly.The expression of miR-141 in cervical cancer was correlated with the clinical stage and serum SCCA level.The expression of lncRNA-H19 in cervical cancer tissue was correlated with lymph node metastasis and serum SCCA level.Pearson analysis showed that the expression levels of miR-141 and lncRNA-H19 in cervical cancer tissues were significantly positively correlated.The overall 3-year survival rate of patients with miR-141 low expression was 37.50%,which was significantly higher than 8.00%of patients with miR-141 high expression.The overall 3-year survival rate of patients with lncRNA-H19 low expression was 41.67%,which was significantly higher than 13.16%of patients with lncRNA-H19 high expression.Conclusion:The expression of miR-141 and lncRNA-H19 in cervical cancer tissues are increased significantly,which were related to the severity of cervical cancer.They may be used as biomarkers for prognosis of cervical cancer.
