槐耳水提取物对三阴性乳腺癌干细胞特性的影响

The Effect of Sophora Japonica Water Extract on the Characteristics of Triple Negative Breast Cancer Metastatic Stem Cells

目的:观察槐耳水提取物对乳腺癌干细胞(CSCs)特性变化的影响。方法:筛选鉴定三阴性乳腺癌CSCs,观察不同浓度槐耳水提取物作用后,YAP/TAZ RNA表达和EMT相关蛋白表达情况,分析每组细胞的差异,定位于细胞核,进行免疫组化分析。同时采用Western blotting、RT-PCR法比较槐耳水提取物处理后CSCs细胞株Hippo通路YAP/TAZ RNA表达情况。结果:槐耳水提取物能抑制三阴性乳腺癌CSCs基因蛋白水平的表达,能抑制YAP/TAZ RNA表达和EMT相关蛋白表达,槐耳水提取物对三阴性乳腺癌CSCs有明显增殖抑制作用,槐耳水提取物能促使CSCs细胞YAP蛋白表达水平及细胞核定位上调,YAP/TAZ RNA表达下调,对CSCs细胞株Hippo通路有明显抑制作用。结论:槐耳水提取物对三阴性乳腺癌CSCs增殖有抑制作用,为中药抗乳腺癌的研究提供了新思路。

Objective:To observe the effect of aqueous extract of Sophora on the characteristics of CSCs.Methods:To screen and identify CSCs of three negative breast cancer,observe the expression of Yap/TAZ RNA and EMT related protein after the action of different concentration of water extract of Sophora,analyze the difference of each group of cells,locate in the nucleus,and carry out immunohistochemistry analysis.At the same time,Western blotting and RT-PCR were used to compare the expression of Yap/TAZ RNA in Hippo pathway of CSCs cell line.Results:The water extract of Sophora can inhibit the expression of CSCs gene protein level,Yap/TAZ RNA expression and EMT related protein expression.The water extract of Sophora can significantly inhibit the proliferation of CSCs in three negative breast cancer.The water extract of Sophora can promote the up regulation of Yap protein expression level and nuclear localization of CSCs cells,The expression of Yap/TAZ RNA was down regulated,which inhibited the Hippo pathway of CSCs cell line.Conclusion:The aqueous extract of Sophora can inhibit the proliferation of CSCs in triple negative breast cancer,which provides a new way for the study of traditional Chinese medicine against breast cancer.
