摘要
目的:对砂珍棘豆总黄酮的提取条件进行优化,并建立紫外-可见分光光度法测定其总黄酮的含量。方法:对影响总黄酮含量的影响因素:提取溶剂、提取方法、浸泡时间、提取时间、取样量进行了考察,以选出最优条件;建立了以芦丁为对照品,采用紫外-可见分光光度法测定砂珍棘豆总黄酮的含量。结果:确定砂珍棘豆中总黄酮的最佳提取条件:用70%甲醇为提取溶剂,浸泡30 min,超声50 min;在此提取条件下,测定5批砂珍棘豆总黄酮的含量为10.13~23.59 mg/g。芦丁在2.5~15.0μg/m L(r=0.999,5)的浓度范围内呈良好的线性关系,平均回收率100.35%,RSD=2.95%。结论:本实验所建立的方法具有操作简单、稳定性好、重现率高等优点,为砂珍棘豆的后续研究奠定了基础。
Objective:To optimize the extraction conditions of total flavonoids from Oxytropis racemosa and establish a method for the determination of total flavonoids by UV-VIS spectrophotometry.Methods:The factors influencing the content of total flavonoids,such as extraction solvent,extraction method,soaking time,extraction time and sampling amount,were investigated to select the optimum conditions.A method for the determination of total flavonoids in Oxytropis racemosa by ultraviolet-visible spectrophotometry with rutin as reference substance was established.Results:The optimum extraction conditions of total flavonoids from Oxytropis vulgaris were determined as follows,using 70%methanol as extraction solvent,soaking for 30 minutes and ultrasonic for 50 minutes.Under this extraction condition,the content of total flavonoids in five batches of Oxytropis racemosa was determined to be 10.13-23.59 mg/g.Rutin showed a good linear relationship in the concentration range of 2.5-15.0μg/mL(r=0.999,5),with an average recovery of 100.35%and RSD=2.95%.Conclusion:The method established in this experiment has the advantages of simple operation,good stability and high reproducibility,which lays a foundation for the follow-up study of Oxytropis racemosa.
