摘要
目的:观察当归芍药散(DSS)活血利水不同配伍对缺血性脑卒中雄性小鼠神经再生的影响,并探讨其对神经再生相关因子—脑源性神经营养因子(BDNF)及趋化因子受体4(CXCR4)的影响。方法:成年C57雄性小鼠,制造大脑中动脉远端梗阻模型(dMCAO模型),随机分为7组,对小鼠进行神经功能评分;通过免疫组化检测SVZ区神经再生,通过Western blot检测对相关指标的影响。结果:与对照组及原方配伍组相比,活血倍量组显著改善了神经功能(P<0.05),增加了DCX阳性细胞数及DCX/BrdU双阳性细胞数。进一步分子机制研究发现,活血倍量配伍显著增加了神经再生相关因子—成熟型BDNF成熟片段的水平及CXCR4的表达(P<0.05,P<0.01)。结论:DSS的活血倍量配伍对缺血性脑损伤小鼠神经再生的贡献度最大。
Objective:To observe the effects of different compatibility of Danggui Shaoyao San(DSS)for activating blood circulation and diuresis on the nerve regeneration of male mice with ischemic stroke,and to explore its effects on brain-derived neurotrophic factor(BDNF)and chemokine receptor 4(CXCR4)these nerve regeneration related factors.Methods:The model of distal middle cerebral artery obstruction(dMCAO model)was established in adult C57 male mice.They were randomly divided into seven groups,and the mice were scored for neurological function.The nerve regeneration in the SVZ region was detected by immunohistochemistry,and the correlation indexes were detected by Western blot method.Results:Compared to the model group and the original formula-compatible group,neural function was significantly improved in the 2x blood activating group(P<0.05);the number of DCX-positive cells and the number of DCX/BrdU double-positive cells increased in the 2x blood activating group;the levels of mature BDNF and the expression of CXCR4 were significantly increased in the 2x blood activating group(P<0.05,P<0.01).Conclusion:The DSS’s blood activating compatibility has the most significant contribution to nerve regeneration in mice with ischemic brain injury.
