降香中1个具有细胞毒活性的异黄烷类新化合物 被引量:4

A new cytotoxic isoflavane from Dalbergiae Odoriferae Lignum

摘要 为研究降香的化学成分,采用硅胶、Sephadex LH-20、高效液相色谱等多种柱色谱技术从降香的乙醇提取物分离得到14个化合物,运用波谱学方法分别鉴定为S-3′-羟基-7,2′,4′-三甲氧基异黄烷(1),2-(2′,4′-二甲氧基苯基)-6-羟基苯并呋喃(2),2-(2′-羟基-4′-甲氧基苯基)-6-甲氧基苯并呋喃(3),7,2′,4′-三甲氧基二氢异黄酮(4),sativanone(5),3,9-二甲氧基-6H-苯并呋喃[3,2-c]色烯-6-酮(6),(6aS,11aS)-homopterocarpin(7),(6aS,11aS)-8-羟基-3,9-二甲氧基紫檀素(8),(6aS,11aS)-3,8,9-三甲氧基紫檀素(9),isodalbergin(10),isoliquiritigenin(11),butein(12),紫铆素(13)和3,7,4′-三羟基黄酮(14),其中化合物1为新化合物,2和3为新天然产物,6,8,9和14为首次从降香中分离得到。采用MTT法测试化合物1~14对人肝癌细胞株BEL-7402、人胃癌细胞株SCG-7901、人肺癌细胞株A549、人慢性髓原白血病细胞株K562和人宫颈癌细胞株HeLa的细胞毒活性。活性测试结果显示化合物1具有显著的细胞毒活性,IC_(50)为2.85~11.62μg·mL^(-1),化合物2,11,12具有弱的细胞毒活性。 Fourteen compounds were isolated from the ethanol extract of Dalbergiae Odoriferae Lignum by various chromatographic techniques,including column chromatographies on silica gel,Sephadex LH-20 and semi-preparative HPLC.Their structures were determined by spectroscopic techniques as S-3'-hydroxy-7,2',4'-trimethoxyisoxane(1),2-(2',4'-dimethoxyphenyl)-6-hydroxybenzofuran(2),2-(2'-hydroxy-4'-methoxyphenyl)-6-methoxybenzofuran(3),7,2',4'-trimethoxydihydroisoflavone(4),sativanone(5),3,9-dimethoxy-6H-benzofuro[3,2-c]chromen-6-one(6),(6aS,11aS)-homopterocarpin(7),(6aS,11aS)-8-hydroxy-3,9-dimethoxypterocarpan(8),(6aS,11aS)-3,8,9-trimethoxypterocarpan(9),isodalbergin(10),isoliquiritigenin(11),butein(12),butin(13)and 3,7,4'-trihydroxyflavone(14).Among them,compound 1 was a new compound,while 2 and 3 were new natural products,6,8,9 and 14 were isolated for the first time from Dalbergiae Odoriferae Lignum.Compounds 1-14 were tested for their cytotoxic activity against human hepatoma cell line BEL-7402,human gastric cancer cell line SCG-7901,human lung cancer cell line A549,human chronic myeloid leukemia cell line K562 and HeLa human cervical cancer cellline by MTT method.Compound 1 exhibited significant cytotoxicity with IC50 values ranging from 2.85 to 11.62μg·mL-1.In addition,2,11 and 12 showed weak cytotoxic activities.
