摘要
目的观察TGF-β1治疗兔SANFH的脂质变化及疗效并探讨其作用机制。方法将44只新西兰大白兔,随机分为对照组(8只)、模型组(18只)、治疗组(18只)。模型组、治疗组均于第2周开始造模。试验期间,对照组正常喂养;模型组:造模后正常喂养;治疗组:造模后肌注TGF-β1并正常喂养。并于造模前、造模中、造模后相对应的时间点对兔血清甘油三酯及总胆固醇含量进行测定,同时于镜下观察双侧股骨头骨组织标本,并计算各组空骨陷窝率。结果第3周末时,与对照组相比,模型组和治疗组的TC、TG、空骨陷窝率均升高(P<0.05);第7周末时,治疗组的TC、TG、空骨陷窝低于模型组,但仍高于对照组(P<0.05)。病理切片和影像学检测均显示模型组出现明显的股骨头坏死表现,而治疗组病理改变较模型组轻。结论表明TGF-β1对血脂有调节作用并对治疗激素性股骨头坏死有确切的疗效。
Objective To observe the lipid changes and efficacy of TGF-β1 in treatment of rabbit SANFH and elaborate the mechanism.Methods A total of 44 New Zealand white rabbits were randomly divided into the control group(n=8),the model group(n=18)and the treatment group(n=18).Both the model group and the treatment group started modeling at the second week.During the experiment,rabbits in the control group were fed normally;rabbits in the model group were fed normally after modeling;those in the treatment group were intramusculally injected with TGF-β1 and normally fed after modeling.At the corresponding time points before,during and after modeling,we measured contents of triglyceride and total cholesterol in rabbit serum,observed the bilateral femoral head tissue specimens under the microscope,and calculated the empty bone lacuna rate.Results By the end of the third week,TC,TG and empty bone lacunae rates were increased in both the model group and the treatment group compared with those in the control group(P<0.05).By the end of the seventh week,TC,TG and empty bone lacuna in the treatment group were lower than those in the model group,but still higher than those in the control group(P<0.05).Pathological sections and imaging examination showed that the model group had obvious necrosis of femoral head,and the treatment group had less pathological change than the model group.Conclusion TGF-β1 has a regulatory effect on blood lipid and is effective in the treatment of steroid induced avascular necrosis of the femoral head.
