金匮肾气丸及六味地黄丸对自然衰老小鼠血浆代谢物的影响 被引量:5

Effect of Jingui Shenqi Pills and Liuwei Dihuang Pills on Metabolic Substances in Plasma of Naturally Aging Mice

摘要 目的比较研究金匮肾气丸及六味地黄丸对自然衰老小鼠血浆代谢物的影响,分析代谢通路富集情况。方法采用基于超高效液相色谱-四级杆-飞行时间质谱(UPLC-Q-TOF/MS)技术的代谢组学方法,经多元统计分析,将质谱同位素分析与数据库检索相结合鉴定衰老相关生物标志物,通过MetPA数据库构建其代谢通路。结果随增龄改变,小鼠体内物质代谢出现紊乱,在筛选的16个与增龄密切相关的代谢物中,金匮肾气丸回调15个物质,六味地黄丸回调14个物质,两方可共同显著回调D-赤藓糖-4-磷酸、二十二碳六烯酸、脱氧羟腐胺赖氨酸、鸟氨酸和16B-羟基四氮唑5个物质,从而影响戊糖磷酸途径、不饱和脂肪酸的生物合成、谷胱甘肽代谢、精氨酸和脯氨酸代谢途径。结论金匮肾气丸及六味地黄丸均可通过影响氨基酸、糖类和脂肪酸代谢来调整自然衰老引起的代谢紊乱。 Objective To compare the effect of Jingui Shenqi Pills and Liuwei Dihuang Pills on metabolic substances in plasma of naturally aging mice;To analyze the enrichment of metabolic pathways.Methods UPLC-Q-TOF/MS-based metabolomics method was used to identify aging-related biomarkers through multivariate statistical analysis,combining mass spectrometry analysis with database search.Metabolic pathways were constructed through MetPA database.Results With aging,the metabolism of substances in mice was disordered.Among the 16 metabolites closely related to aging,Jingui Shenqi Pills had different degrees of callback to 15 substances,and Liuwei Dihuang Pills had different degrees of callback to 14 substances,and both could significantly callback D-Erythrose 4-phosphate,docosahexaenoic acid,deoxyhypusine lysine,ornithine,and 16b-hydroxystanozolol,thereby affecting pentose phosphate pathway,biosynthesis of unsaturated fatty acids,glutathione metabolism,arginine and proline metabolism pathway.Conclusion Both Jingui Shenqi Pills and Liuwei Dihuang Pills can adjust the metabolic disorder caused by natural aging by affecting the metabolism of amino acids,sugars and fatty acids.
