新型单面刻槽药物洗脱支架的制备与研究

摘要 为研究单面刻槽支架表面载药涂层的制备,采用压电微喷技术制备以聚乳酸(PLA)为载药物的雷帕霉素药物洗脱支架。通过调节影响压电微喷技术的主要参数,使涂层效果达到最优,并对支架表面的凹槽进行扫描和识别,以制备药物洗脱支架。利用球囊扩张实验来检验载药涂层与支架基体之间的结合情况,并在扫描电子显微镜(SEM)下观测扩张前后药物洗脱支架表面涂层的形貌特征;在PBS缓冲液中对制备的药物洗脱支架进行体外模拟实验,并绘制药物释放的动力学曲线。结果表明:压电微喷技术应用于单面刻槽支架表面载药涂层制备具有可行性,能实现对凹槽的精准喷涂,并且经球囊扩张后的支架表面涂层牢固度达到要求,药物释放曲线也能够满足释放要求。 In order to study the preparation of drug-loading coating on the surface of abluminal grooves stent,rapamycin loaded PLA eluting stent was prepared by piezoelectric microinjection technique.The coating effect was optimized by adjusting the main parameters of piezoelectric microinjection technology,and the grooves on the surface of the stent were scanned and identified to prepare drug-eluting stents.Balloon dilatation experiment was used to evaluate the combination between drug-loading coating and scaffold substrate,and the morphology of drug-eluting stent surface coating before and after dilatation was observed under scanning electron microscope(SEM).The in vitro release behavior of drug-eluting stent was characterized assessed in PBS buffer,and the kinetics curve of drug release was plotted.The results showed that the application of piezoelectric micro-jet technology in the preparation of drug-loading coating on the surface of abluminal grooves stent was feasible,and the precision spraying of the grooves could be achieved.Moreover,the firmness of coating on the surface of the stent after balloon expansion met the requirements,and the drug release curve could also meet the release requirements.
