基于营养液静置培养方式的蓝莓试管苗增殖研究

摘要 [目的]提高蓝莓品种斯巴坦试管苗的增殖效率。[方法]以蓝莓斯巴坦试管苗为材料,采用正交试验设计,研究了不同支撑物(琼脂、珍珠岩、蛭石)、WPM营养液浓度(WPM、1/2WPM、1/4WPM)以及ZT的不同浓度(0.5、1.0、1.2 mg/L)对蓝莓试管苗增殖的影响。[结果]通过正交试验确定3个因子对蓝莓试管苗增殖的影响顺序为:支撑物>营养液浓度>ZT浓度。[结论]蓝莓丛生芽增殖最适宜的营养液静置培养方式为珍珠岩+WPM+1 mg/L ZT,为蓝莓斯巴坦工程化育苗提供了技术基础。 [Objective]The objective of this study was to improve the efficient of proliferation of tissue culture seeding of Vaccinium uliginosum‘Spartan’in vitro.[Method]The seedlings of Vaccinium uliginosum‘Spartan’in vitro were taken as experimental materials,and the effects of the different supports(agar,perlite,vermiculite),different concentrations of nutrient solution(WPM,1/2 WPM and 1/4 WPM),and different concentrations of ZT(0.5,1.0 and 1.2 mg/L)on proliferation of blueberry seedlings in vitro were studied.[Result]The effect of the three factors for proliferation culture was in the order of supports>nutrient solution concentration>concentrations of ZT.[Conclusion]The optimizing medium for proliferation culture was perlite+WPM+1 mg/L ZT.The result appeared to provide a basis for the Vaccinium uliginosum‘Spartan’of the seedling propagation.
