自血疗法对慢阻肺稳定期模型大鼠炎性反应相关指标的影响

Effects of Autohemotherapy on Indexes of Inflammatory Response in Rats with COPD in Stable Phase

目的:研究自血疗法对慢阻肺稳定期模型大鼠白细胞介素-2(IL-2)、IL-6及C反应蛋白(CRP)、降钙素原(PCT)水平的影响。方法:选取60只健康清洁级雄性Wistar大鼠作为研究对象,将其以随机抽签法等分成空白对照组、空白观察组以及自血疗法组。其中空白观察组及自血疗法组大鼠均由熏香烟联合气道内灌注毒素法制作慢阻肺稳定期模型。完成后予以空白对照组、空白观察组大鼠生理盐水灌服,自血疗法组大鼠则予以自血疗法干预。3组大鼠均进行30 d治疗,比较治疗前后血清IL-2、IL-6及CRP、PCT,血清基质金属蛋白酶-9(MMP-9)以及金属蛋白酶抑制剂1(TIMP-1)变化情况。结果:治疗后自血疗法组大鼠IL-2、IL-6及CRP、PCT均低于空白观察组,但高于空白对照组(均P<0.05)。治疗后自血疗法组大鼠血清MMP-9以及TIMP-1均低于空白观察组,但高于空白对照组(均P<0.05)。结论:自血疗法可显著改善慢阻肺稳定期模型大鼠的炎性反应程度,降低血清MMP-9以及TIMP-1水平,可能成为临床治疗慢阻肺稳定期的有效手段之一。

Objective:To analyze the effects of autohemotherapy on the levels of interleukin-2(IL-2),IL-6,c-reactive protein(CRP)and procalcitonin(PCT)in rats with chronic obstructive pulmonary disease(COPD)in stable phase.Methods:A total of 60 healthy and clean male Wistar rats were selected and divided into blank control group,blank treatment group and autohemotherapy group by random lottery.Rats in the blank treatment group and the autohemotherapy group were modeled by cigarette smoking combined with intraairway toxin infusion.After modeling,the rats in the blank control group and the blank treatment group were drenched with normal saline,and the rats in the autohemotherapy group were given the autohemotherapy intervention.All rats in the three groups were treated for 30 days,the changes of serum IL-2,IL-6,CRP,PCT,serum matrix metalloproteinase-9(MMP-9)and metalloproteinase inhibitor 1(TIMP-1)before and after treatment were compared.Results:After treatment,the levels of IL-2,IL-6,CRP and PCT in the autohemotherapy group were lower than those in the blank treatment group,but was higher than the blank control group(all P<0.05).After treatment,serum levels of MMP-9 and TIMP-1 in rats from the autohemotherapy group were lower than those in the blank treatment group,but was higher than the blank control group(all P<0.05).Conclusion:Autohemotherapy can significantly alleviate the inflammatory response,reduce serum MMP-9 and TIMP-1 of the model rats.It may be one of the effective methods to treat chronic obstructive pulmonary disease in stable phase.
