摘要
目的:建立HPLC波长切换法同时测定甲氯霜中甲硝唑、氯霉素和羟苯乙酯的含量。方法:采用Wondasil C18色谱柱(250 mm×4.6 mm,5μm),以甲醇(流动相A)-0.5%冰乙酸溶液(流动相B)为流动相,梯度洗脱(0~8 min 35%A;8~16 min 35%~45%A,16~30 min 45%~60%A);检测波长为320 nm(0~8 min)、278 nm(8~16 min)、254 nm(16~30 min);柱温为40℃;流速为1.0 ml/min。结果:甲硝唑、氯霉素和羟苯乙酯分别在38.68~77.36μg/ml(r=0.9999)、38.24~76.49μg/ml(r=0.9999)和1.87~3.75μg/ml(r=0.9999)范围内线性关系良好,平均回收率分别为(99.4±0.4)%、(99.3±0.8)%和(98.9%±0.4)%(n=9)。结论:本研究所建立的HPLC波长切换法简便、快速、准确、重复性好,适用于甲氯霜中3种成分的同时测定。
Objective:To establish a HPLC wavelength switching method for simultaneous determination of the components of metronidazole,chloramphenicol and ethylparaben in Jialv cream.Methods:The Wondasil C18 column(250 mm×4.6 mm,5μm)was adopted with the mobile phase consisting of methanol(phase A)and 0.5%glacial acetic acid(phase B),gradient elution(0-8 min 35%phase A,8-16 min 35%-45%phase A,16-30 min 45%-60%phase A).The detection wavelengths were 320 nm(0-8 min),278 nm(8-16 min)and 254 nm(16-30 min).The column temperature was set at 40℃.The flow rate was 1.0 ml/min.Results:The linear ranges of metronidazole,chloramphenicol and ethylparaben were 38.68-77.36μg/ml(r=0.9999),38.24-76.49μg/ml(r=0.9999)and 1.87-3.75μg/ml(r=0.9999),respectively.The average recoveries of the three components were(99.4±0.4)%,(99.3±0.8)%and(98.9±0.4)%(n=9).Conclusion:The established HPLC wavelength switching method is simple,rapid,accurate and reproducible,which is suitable for simultaneous determination of three components in Jialv cream.
