AFLP银染法和荧光标记法对文蛤遗传多样性评价的效果分析 被引量:1

摘要 以文蛤山东种群(SD)和广西种群(GX)为实验材料,比较分析了AFLP银染法和荧光标记法对遗传多样性的评价效果。结果显示,AFLP荧光标记法在选择性扩增引物5’端加了FAM荧光标记,产物直接在DNA测序仪上检测,自动采集图像和数据,较银染法的数据准确度更高、稳定性更好。从遗传参数来看,银染法和荧光标记法获得的GX种群的多态位点比例、Nei’s基因多样性指数和香侬信息指数均高于SD种群,表明两种方法遗传多样性评价的结果一致。AFLP荧光标记法在文蛤两种群中共获得244个扩增位点,多态位点比例为94.26%,均高于银染法(177个扩增位点,多态位点比例85.31%)。文蛤SD和GX种群的扩增位点数、多态位点比例、Nei’s基因多样性指数和香侬信息指数均是荧光标记法>银染法。 To compare the effects of AFLP silver staining and fluorescent marker on the evaluation of biological genetic diversity,Shandong population(SD)and Guangxi population(GX)in Meretrix meretrix were analyzed.The results showed that the method of AFLP fluorescence marker is more efficient,accurate and stable than the silver staining method,because of adding FAM fluorescence marker to the 5'end of the selective amplification primers,the detection of PCR products by DNA sequencer,and the automatic collection of the electrophoretogram and data.In terms of genetic parameters,the percentage of polymorphic loci,Nei's genetic diversity index and Shannon genetic information index of GX population obtained by AFLP silver staining and fluorescent marker were all higher than that of SD population,indicating that the evaluation results of genetic diversity by using two methods were consistent.A total of 244 amplification loci were obtained by AFLP fluorescence marker in both populations,with the polymorphic loci percentage of 94.26%,both higher than that by silver staining(177 amplification loci,polymorphic loci percentage of 85.31%).The number of amplification loci,percentage of polymorphic loci,Nei's genetic diversity index and Shannon genetic information index of SD and GX populations by fluorescent marker were all more than those by silver staining.
