Ferroptosis is an iron-dependent form of regulated cell death caused by accumulation of lipid-based reactive oxygen species.The initiation and execution of ferroptosis lie at the intersection of iron homeostasis,lipid metabolism and antioxidant system.Based on the insight into the cross-talk between ferroptosis-and cancer-associated pathway,a series of ferroptosis-inducing agents have been designed and developed for cancer cells,including experimental compounds,clinical drugs and nanomaterials.This review summarized the regulatory mechanisms of ferroptosis and the strategies of cancer therapy based on ferroptosis.
