当归多糖对头孢噻呋钠联合LPS致鸡肝损伤的防治效果 被引量:1

Prevention and Treatment Effects of Unprocessed Angelica sinensis Polysaccharides on Liver Injury Induced by Ceftiofur Sodium Combined with LPS

摘要 探讨当归多糖(Unprocessed Angelica sinensis polysaccharide,UASP)对头孢噻呋钠联合内毒素(lipopolysaccharide,LPS)致鸡肝损伤的防治效果,为家禽保肝药物研发奠定基础。基于前期试验研究,购买1日龄健康海蓝褐蛋鸡264羽,首先开展头孢噻呋钠联合不同来源LPS致鸡肝损伤对比试验研究、头孢噻呋钠联合脂多糖粗提液(LPS I)致鸡肝损伤时间点与剂量的重复性筛选研究,其次开展UASP对头孢噻呋钠联合LPS I致鸡肝损伤的防治效果研究。主要评价指标为鸡临床体征、丙氨酸氨基转移酶(ALT)与天门冬氨酸氨基转移酶AST)及肝脏病理组织学观察。头孢噻呋钠联合不同来源LPS致鸡肝损伤对比研究结果显示,各模型组鸡血清AST和ALT均显著升高(P<0.05),结合临床症状与肝脏组织观察,头孢噻呋钠联合LPS I可以造成显著肝损伤;头孢噻呋钠联合LPSⅠ致鸡肝损伤时间点与剂量的重复性筛选研究显示,固定头孢噻呋钠剂量为5 mg/kg/d,LPSⅠ剂量为4 mL/kg时,造模后6 h与48 h肝损伤均较为明显;UASP干预头孢噻呋钠联合LPS I致鸡肝损伤的效果研究显示,模型组鸡血清AST和ALT较正常对照组显著升高(P<0.05),与模型组相比,UASP高剂量组(UASPH)血清AST和ALT均显著降低(P<0.05)。显微镜下显示,模型组鸡肝细胞出现广泛变性、坏死;与模型组相比,UASP组肝细胞变性坏死减轻,UASPH组改善更明显。说明UASP对头孢噻呋钠联合LPS诱导的肝脏损伤具有保护作用。 To investigate the preventive and therapeutic effects of unprocessed Angelica sinensis polysaccharide(UASP)on liver injury induced by ceftiofur sodium(CS)combined with Lipopolysaccharide(LPS)in chickens,and to lay a foundation for the research and development of liver-protecting drugs for poultry,264 healthy Hai-lan-he chickens of one-day-old were purchased based on the preliminary experimental study.Firstly,the comparative study of liver injury induced by CS combined with LPS from different sources,the repetitive screening study of the time points and doses of liver injury induced by CS combined with homemade LPS(LPS I)were carried out.Secondly,the preventive and therapeutic effects of UASP on liver injury induced by CS combined with LPS I were studied.The main evaluation indicators include the clinical symptoms of chickens,indexes of alanine transaminase(ALT)and aspartate aminotransferase(AST),and the observation of liver histopathology.The results of comparative study on liver injury induced by CS combined with LPS from different sources showed that AST and ALT in sera of chickens in each model group increased significantly(P<0.05),and combined with clinical symptoms and liver tissue observation result,the combination of CS and LPS I could cause significant liver injury.Repeated screening study on time point and dose of chicken liver injury induced by homemade LPS showed that the liver injury was more obvious at 6 h and 48 h when fixed dose of CS as 5 mg/kg/d and LPS dose as 4 mL/kg;The results of UASP on the liver injury induced by CS combined with LPS I in chickens showed that the levels of AST and ALT in chicken serum in model group were significantly higher(P<0.05)than that in K group.Compared with M group,the levels of AST and ALT in chicken serum in UASPH group were significantly lower(P<0.05).Microscopically,extensive degeneration and necrosis of chicken hepatocytes in M group appeared.Compared with M group,the degeneration and necrosis of hepatocytes were alleviated in the UASP groups,and the improvement was more obvious in UASPH group.The UASP has a protective effect on the liver injury induced by CS combined with LPS.
