摘要 目的研究家蝇抗菌蛋白对小鼠皮肤烫伤模型创伤愈合的影响。方法将36只健康成年小鼠随机分为生理盐水组、磺胺嘧啶银组和抗菌蛋白组,制备皮肤烫伤模型,将供试品按0.2 mL/只的剂量涂抹创面上,1次/d。于损伤后第7、14、21天测量皮肤损伤面积,计算伤口愈合率,同时用免疫组化试剂盒常规方法测BFGF、EGF含量。第21天取损伤部位皮肤,组织切片观察其形态学变化。结果第21天抗菌蛋白组和磺胺嘧啶银组小鼠皮肤损伤部位愈合率均值分别为93.15%和91.79%,与生理盐水组均值71.32%比较,提高了21.83%和20.47%(P<0.05);组织形态学显示,21 d时抗菌蛋白组皮肤组织已完全愈合,对照组尚未完全愈合。抗菌蛋白组BFGF及EGF表达在14、21 d明显超过对照组,差异有显著性(P<0.05)。结论家蝇抗菌蛋白能改善小鼠烫伤部位肉芽组织生长状态,可能是通过促进BFGF、EGF的表达,从而促进创面的修复过程。 Objective To study the effect of antibacterial protein from Musca domestica larvae on wound healing in mouse skin scald model.Methods 36 healthy adult mice were randomly divided into normal saline control group,topical sulfadiazine silver group,and Musca domestica larvae antibacterial protein group.The skin scald model was prepared.The test product was applied to the wound surface at a dose of 0.2 mL/mouse,once a day.The skin injury area was measured on day 7,14,and 21 after injury,and the wound healing rate was calculated.At the same time,the contents of BFGF and EGF were measured by conventional methods of Y immunohistochemistry kit.On the 21st day,the skin of the injured site was taken,and the morphological changes were observed in tissue sections.Results On the 21st day,the average healing rates of skin burn in mice in the antibacterial protein group and the silver sulfadiazine group were 93.15%and 91.79%,respectively,which were 21.83%and 20.47%higher than the mean of 71.32%in the normal saline control group(P<0.05).Histomorphology showed that the skin tissue of the antibacterial protein group had completely healed on the 21st day,while the control group had not completely healed.The expressions of BFGF and EGF in the antibacterial protein from Musca domestica group were significantly higher than those in the control group on day 14 and 21,and the differences were significant(P<0.05).Conclusion Antibacterial protein from Musca domestica larvae can improve the growth state of granulation tissue in burns of mice,which may promote the repair process of wounds by promoting the expression of BFGF and EGF.
