摘要
目的:探索提高间充质干细胞(mesenchymal stem cells,MSCs)表面趋化因子受体4(CXCR4)表达的方法。方法:分别对兔骨髓间充质干细胞(Rabbit-MSCs)和人脐带间充质干细胞(HUC-MSCs)进行培养,采用3%低氧环境、不同浓度基质细胞衍生因子-1(SDF-1)(1 ng/mL,10 ng/mL,100 ng/mL)进行预处理,24 h后采用CCK-8试剂盒检测细胞活性,流式细胞仪及免疫荧光染色检测MSCs表面CXCR4表达情况。结果:两种预处理方式对MSCs活性无影响,经低氧和SDF-1预处理后MSCs表面CXCR4表达提高,低氧处理较SDF-1效果更好。结论:低氧环境对提高间充质干细胞表面CXCR4表达具有更好的效果。
Objective:To increase the expression of CXCR4 on the surface of mesenchymal stem cells.Methods:Rabbit bone marrow derived from mesenchymal stem cells(rabbit-MSCs)and human umbilical cord derived from mesenchymal stem cells(HUC-MSCs)were cultured for 48 hours and then treated with hypoxia condition and SDF-1(with concentration of 1 ng/mL,10 ng/mL,100 ng/mL)for 24 hours.The viability of MSCs was evaluated by cell counting kit-8(CCK-8).The expression of CXCR4 on the surface of MSCs was detected by flow cytometry and immunofluorescence staining.Results:The results of the CCK-8 showed that the MSCs activity was not affected by treatment.There is a certain CXCR4 expression improvement by hypoxia treatment and SDF-1(1 ng/mL,10 ng/mL,and 100 ng/mL)compared with normal MSCs.Furthermore,hypoxia treatment showed better effects than SDF-1.Conclusions:Hypoxic environment has a good effect on increasing CXCR4 expression on the surface of mesenchymal stem cells.
