响应面法优化产细菌纤维素菌发酵条件 被引量:12

Optimization of fermentation conditions for enhanced bacterial cellulose production by using response surface method

摘要 为了提高细菌纤维素(bacterial cellulose,BC)的产量,以本实验室筛选出的产BC菌Komagataeibacter rhaeticus TJPU03(K.rhaeticus TJPU03)作为研究对象,通过响应面法设计实验优化BC的生产条件。将优化内容分为两部分:培养基组分和外部因素。利用软件Design expect 8.0计算和优化BC的最佳生产条件。结果表明:当外部条件为温度28.52℃、初始pH值3.0、接种量8.87 mL以及发酵培养基组分为葡萄糖1.58%、酵母粉0.66%、蛋白胨0.42%、柠檬酸0.12%、磷酸氢二钠0.3%时,实际BC产量可达6.7432 g/L,较未优化前显著提高。 In order to increase the yields of bacterial cellulose(BC),the new BC-producing strain Komagataeibacter rhaeticus TJPU03(K.rhaeticus TJPU03)screening in our laboratory was chosen to explore the high-production condition of BC by response surface method(RSM).The optimization included fermentation medium components and external condition.Design expect 8.0 was used to design the experiment and calculate the optimal conditions.The results showed that the actual production of BC could reach 6.7432 g/L at the optimal fermentation medium components(glucose 1.58%,yeast paste 0.66%,peptone 0.42%,citric acid 0.12%and disodium hydrogen phosphate 0.3%)and optimal external condition(temperature of 28.52℃,initial pH value of 3,and inoculation amount of 8.87 mL).The production of BC was improved obviously after optimization.
