冬凌草甲素对角质形成细胞增殖、凋亡、迁移的影响 被引量:5

摘要 目的 研究冬凌草甲素(oridonin)对人永生化角质形成细胞株(HaCaT cells)增殖、凋亡和迁移的影响,并分析其机制。方法 分别用0、10、20和40 μmol/L的冬凌草甲素处理HaCaT细胞,在不同时间用MTT法检测冬凌草甲素对HaCaT细胞增殖的抑制作用;用流式细胞仪检测冬凌草甲素对HaCaT细胞凋亡的影响;用Transwell实验检测冬凌草甲素对HaCaT细胞迁移能力的影响;用ELISA实验检测冬凌草甲素对HaCaT细胞分泌肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)水平的影响。结果 冬凌草甲素对 HaCaT细胞的增殖有明显的抑制作用,且抑制率与浓度和时间呈正相关;经冬凌草甲素干预后的HaCaT细胞凋亡率升高,迁移减慢;冬凌草甲素作用于HaCaT细胞后TNF-α、IL-6表达水平下降。结论 冬凌草甲素能抑制角质形成细胞的增殖、迁移,促进凋亡,并且可以抑制角质形成细胞TNF-α、IL-6的分泌,可能成为治疗银屑病的潜在药物。 Objective To study the effects of oridonin on the proliferation,apoptosis and migration of HaCaT cells and analyze its mechanism.Methods HaCaT cells were treated with 0,10,20 and 40 μmol/L of oridonin,and the proliferation inhibition effect of oridonin on keratinocytes was detected by MTT at different times;flow cytometry was used to detect the apoptosis;Transwell assay was used to detect the migration of keratinocytes;ELISA was used to detect tumor necrosis factor(TNF-α)and interleukin-6(IL-6)secreted by HaCaT cells.Results The cell proliferation of HaCaT cells treated with oridonin was significantly decreased,and it was positively correlated with concentration and time.The apoptosis rate of HaCaT cells increased after oridonin intervention,and the migration slowed down;the expression levels of TNF-α and IL-6 decreased after treatment with oridonin in HaCaT cells.Conclusion Oridonin can inhibit the proliferation and migration of keratinocytes,promote apoptosis,and inhibit the secretion of TNF-α and IL-6 from keratinocytes,which may become a potential drug for the treatment of psoriasis.
